    • 功能

      DNA repair enzyme that incises DNA at 8-oxoG residues. Excises 7,8-dihydro-8-oxoguanine and 2,6-diamino-4-hydroxy-5-N-methylformamidopyrimidine (FAPY) from damaged DNA. Has a beta-lyase activity that nicks DNA 3' to the lesion.

    Use of ab98249 - Recombinant Human Ogg1 protein for comet assay

    We used hOGG1 for the enzyme-modified comet assay, in order to detect oxidative DNA damage.
    We used human peripheral blood mononuclear cells (PBMC) and treated them with 3mM potassium bromate to induce oxidative DNA damage.
    We tested a range of enzyme dilutions, from 1:10 to 1:100,000, and tested incubation times from 10 minutes to 2 hours (although 15 minutes should be enough according to our previous experience with this enzyme from other brands).
    Notable increases in oxidative DNA damage were only observed after 1 and 2 hours of incubation, and using low enzyme dilutions (1:500 or less). In addition, the net OGG1 sensitive sites observed did not exceed 20%, that is a very low value considering the potassium bromate concentration used to induce damage.
    In conclusion, the enzyme concentration to be used for detecting a positive response in the comet assay is too high to be economically feasible, at least in the version of the comet assay using two gels per slide, and especially for human biomonitoring studies.
