This antibody produced an intense staining in the whole brain, while used 1:3000 diluted. The staining is so strong that it is difficult in some areas to distinguish in which cell subtype it is localized. It is particularly true in areas rich in synapses, such as dendrate girus. The pictures loaded show the staining obtained in the dentate girus at low (A) and high (B) power magnification.
The sections used came from animals perfused fixed with Paraformaldehyde 4% with 15% of a solution of saturated picric acid, in phosphate buffer 0.1M. Following postfixation in the same fixative overnight, the brains were cryoprotected in sucrose 30% overnight. Brains were then cut using a cryostat and the immunostainings were performed using the ‘free floating’ technique.
