      Immunohistochemical analysis of dog skin tissue, detecting staining of NQO1 with ab98784.

      Tissue was fixed with formaldehyde and blocked with none for ; antigen retrieval was by heat mediation in a citrate buffer (pH 6). Samples were incubated with primary antibody (ab28947 Mouse monoclonal [A180] to NQO1 at 1/175 in BSA in TBS) for 45 minutes. ab98784 (1/500) was used as the secondary antibody.

    16 weeks old Sprauge Dawley knee samples. Decalcified with Immunocal for 24h. Frozen sectioned to 8μm. Fixation: Methanol w/ 3% hydrogen peroxide. Blockage: 1.5% Rabbit Serum (Abcam, ab7487). Primary: Mouse Anti-Type II Collagen (Chondrex, Cat. 7048, 1:1000). Secondary: Rabbit Anti-Mouse IgG H&L (Biotin) preadsorbed (Abcam, ab98784, 1:1000). ABC HRP Kit (Vector Laboratories, PK-4002). DAB Peroxidase Substrate Kit (Vector Laboratories, SK-4100)

    Thank you very much for your reply.
    I did vary goat anti-mouse IgG concentration, 1:25, 1:50, 1:100. In
    all cases, even without primary antibody, I do see signals in
    controls. Here are the conditions used:
    goat anti-mouse IgG: various conc
    Primary: Mouse anti-CD68 [ED1] (Abcam ab31630) (or isotype or none)
    Secondary: Rabbit anti-mouse IgG-biotin (Abcam ab98784)
    HRP: Streptavidin-peroxidase polymer, ultrasensitive (Sigma S2438)
    I am not 100% aware of what my supervisor has told you (as I joined
    the team in the middle of this project and inherited the project from
    a student. I myself have not tried FA-11 from Serotec in our tissue
    of interest, kidney, but it did work for lung when I tried. Also, in
    general, Abcam antibodies are of higher quality. So I have high hope
    that FA-11 from Abcam can detect macrophage infiltration.

    Since the no primary control is showing staining, it's probably an issue with the secondary antibody. But I'll be happy to send you ab53444 as a free of charge replacement for the CD68 antibody. You'll have to use an anti-rat secondary antibody with ab53444 and I strongly encourage you to use one that is pre-adsorbed against mouse so you don't run into a similar problem.

    I am sorry this product did not perform as stated on the datasheet and for the inconvenience this has caused. As requested, I have issued a free of charge replacement of ab53444.
