This is strange, if the red cells are seen even without primary antibody then the problem is either due to secondary antibody or due to protocol. I have checked the protocol and I find no problem with it other than the wash step which I assume is 3 washes of 5 minutes.
You may need to try again, following the correct ICC/IF protocol steps.

- What was the cell line used? e.g. human liver cancer cell line.
- Do you see difference in no primary control? Were the cells red in colour as seen with primary antibody?
- Do you think the problem is due to ab6718 or ab110641.
- What was the storage condition of antibody?
- Did you change any step when doing repeat experiment?

I am very sorry to hear you are still experiencing problems with these products. According to the images sent it appears the secondary is still giving a lot of non specific background.

If the antibody is still under the Abpromise guarantee I will be more than happy to send you a free of charge replacement for a different Goat anti-Rb secondary.

In regards to the primary anti-CD31, I am afraid this antibody has not been tested and validated in ICC, and therefore we cannot guarantee it will work in this application.

I would suggest trying the new secondary I will send you free of charge, and in case you are still not obtaining the expected results, we will study more carefully the protocol and different possibilities.

What is the isotype of the primary antibody? Has it been validated to work in ICC?

Thank you for your enquiry. 1) Ab27089 is supplier as a pre diluted product and Ab4055 is not. 2) We do not have specific feedback in relation to certain secondary Abs functioning in conjunction with our antibodies; however a list of compatible secondary Abs are cited at the base of each online data sheet for you to choose from. I would expect any non rabbit species anti rabbit IgG with a suitable conjugation to function. 3)Looking at products Ab7087 and Ab6718 the only obvious difference that I am able to spot is the different concentrations of the product. I will pass this information onto my colleague sin business development since it is possible that we have added a duplicate product in error. Thank you for your enquiry. According to the datasheet this antibody is in liquid form (the buffer is: 0.02M K Phosphate, 0.15M NaCl, 0.01% Sodium Azide, pH7.2) so you do not need to reconstitute it.
