      Flow Cytometry - Goat Anti-Mouse IgG - H&L (SureLight® APC), pre-adsorbed (ab99900)
      Fluorescence excitation and emission spectra of Allophycocyanin in 100mM sodium phosphate (pH7.2) + 1mM EDTA and 1mM sodium azide. The emission scan was taken with excitation at 630nm. The excitation scan was taken with an emission at 660nm. The curves were normailized to equalize peak heights.

    This secondary antibody has only been tested for Flow cytometry. However, as long as it is compatible with the primary antibody (host species, isotype), it should be fine for immunofluorescence microscopyas well.

    Inquiry: Sehr geehrter Support Service, bezüglich einer durchflusszytometrischen Methode trat ein Problem mit der Antikörperkopplung auf. Folgende Antikörper wurden verwendet: Anti-Digoxigenin antibody [BT.21H8] (Biotin) (ab419) Goat polyclonal Secondary Antibody to Mouse IgG - H&L (SureLight Allophycocyanin), pre-absobed (ab99900)
    Das Experiment wird in folgenden Versuchsansätzen durchgeführt:
    1.Versuch: Zunächst erfolgt die Kopplung eines Oligos an einem Bead. Das Oligo enthält an seinem freien nicht gebundenen Ende das Digoxigenin. Dies stellt die Bindungsstelle für den Anti-Digoxigenin antibody [BT.21H8] (Biotin) (ab419) dar. Dieser bindet. Im letzten Schritt bindet über das Biotin am Antikörper ein Floureszenzfarbstoff. Es erfolgt die Messung am Durchflusszytometer und es kann bestätigt werden, dass die Kopplung funktioniert.
    2.Versuch Zunächst erfolgt die Kopplung eines Oligos an einem Bead. Das Oligo enthält an seinem freien nicht gebundenen Ende das Digoxigenin. Dies stellt die Bindungsstelle für den Anti-Digoxigenin antibody [BT.21H8] (Biotin) (ab419) dar. Dieser bindet. Im letzten Schritt sollte der Antikörper Goat polyclonal Secondary Antibody to Mouse IgG - H&L (SureLight Allophycocyanin), pre-absobed (ab99900) binden, welche die abschließende Detektion mittels Durchflusszytometer durch das Allophycocyanin ermöglicht.
    Kopplung funktioniert nicht. Detektion ist nicht möglich.
    Meine Fragen an Sie wären folgende:
    Können diese zwei Antikörper miteinander binden? Wo befindet sich das Biotin am ersten Antikörper? Stört Biotin die Bindung des Sekundärantikörpers am Primärantikörper? Wäre es ratsam den 2.Versuch mit einem Primärantikörper durchzuführen, welcher kein Biotion enthält? Über eine baldige Rückmeldung bin ich Ihnen sehr dankbar.

    Vielen Dank für Ihre Anfrage.

    Wenn Antikörper mit Biotin gekoppelt werden, wird das meist über freie Anime getan. Das bedeutet das Biotin über den ganzen Antikörper verteilt ist. Die Antikörper werden dann darauf getestet, dass die Bindestelle, die das Zielprotein bindet, nicht von Biotin besetzt ist.

    Lange Rede kurzer Sinn: Die Biotin-Moleküle stören sehr wahrscheinlich wenn dann ein Zweitantikörper eingesetzt wird um den primären Antikörper zu binden.

    Ich schlage deshalb, wie Sie schon richtig vermuteten, vor einen ungekoppelten primaeren Antikörper für Versuch 2 zu verwenden.

    Um den unwahrscheinlichen Fall auszuschließen, dass es sich bei dem sekundären Antikörper um einen fehlerhaften Antikörper handelt, schlage ich aber vor, zuerst den sekundären mit einem anderen primären zu testen. Falls der sekundäre einen anderen angekoppelten primären Antikörper aus der Maus nicht erkennt, werde ich Ihnen den sekundären Antikörper gerne ersetzen.

    Ich hoffe, ich konnte Ihnen mit diesen Informationen weiterhelfen. Bitte lassen Sie mich wissen, falls Sie weitere Fragen haben oder der sekundäre Antikörper im Test mit einem un-gekoppelten primären Antiköper nicht funktioniert.

    Question

    Thanks for your reply and troubleshooting questionnaire. Today I managed
    to get some signal in florescence microscope - and most likely the problem
    earlier was antibody aggregation, as I centrifuge the antibody aliquot at 4
    degree and used the supernatant, and it worked well with the recommended
    concentration.
    I'll also try the FACS again sometime during next week or so, and if
    have any further problem will contact you. You can also find the answers for
    all your questions below, in case you find some other point where I should
    pay attention.

    Question

    Hi Technical Support Team,
    I have an antibody for FACS Goat polyclonal Secondary AB to mouse
    IgG-H (Sure Light Allophycocynin), per-adsorbed, code: ab99900. As
    recommended in the datasheet, I reconstituted the antibody in 1 ml of ddH2O
    and then stored at 4deg C. Now when I'm trying to use this antibody for cell
    sorting, it did not show any signal. I have also tried using higher
    concentration of antibody without any improvement. Kindly suggest.

    In order to understand the problem better and to try to find the origin of the problem, I appreciate if you could confirm the following details. These details will enable us to investigate this case and will provide us with vital information for monitoring product quality.

    1.) Do you know whether the primary antibody used is working? Have you tested it with another secondary antibody?

    2.) Can you please describe what species and isotype the primary antibody is from?

    3.) Can you please provide also details of the dilutions used of the secondary antibody.

    4.) Have the filters of the machine be checked to be able to read the APC fluorochrome? What laser and filter have been used?

    5.) Howhas the antibody beenstored?

    6.)How many times haveyou tested this antibody in total? Have you obtained no results each time?

    7.) Finally, can you please also provide the order number of the purchase or the approximate order date with the name of the person ordering? This will help me to locate the order and the vial with its batch number.
