Alpaca monoclonal [14A4] Anti-Mouse IgG2a/b antibody (AZdye 568) (ab325298) is a ready-to-go fluorescent camelid single-domain antibody (sdAb, nanobody, VHH) conjugate with maximum absorption wavelength of 578 nm and a maximum emission wavelength of 602 nm. Ideal for fluorescent cell and tissue imaging. Suitable for ICC/IF applications.

Immunocytochemistry/ Immunofluorescence - Alpaca monoclonal [14A4] Anti-Mouse IgG2a/b antibody (AZdye 568) (AB325298)
Immunofluorescent analysis of 4% paraformaldehyde fixed, 0.1% TritonX-100 permeabilized, quenched with 0.1M ammonium chloride, and blocked with 10% normal goat serum in PBS HeLa (human cervical cancer cells) cells labelling beta Tubulin with ab315214 at 1 : 2000 (0.50 µg/ml) dilution, followed by ab325298 Alpaca monoclonal [14A4] Anti-Mouse IgG2a/b antibody (AZdye 568) at 1/500 (0.15 µg/ml per sdAb) dilution (red).

Image was taken with a fluorescence microscope (Zeiss Axio Imager Z1).
ab92742 anti-Ki67 antibody was used to counterstain Ki67 at 1/1000 (0.85 µg/ml) dilution (yellow).

Together with their monovalent binding mode, the defined fluorophore conjugation of ab325298 results in a quantitative and reliable readout for several fluorescence-based applications. It contains a defined number of fluorophores at known positions. In addition due to the small size the fluorophore displacement from the bound target protein is very low making ab325298 ideal for standard super resolution technologies.

- High affinity monovalent binding + two fluorophores per sdAb for brighter signals.
- Reagent is made from small-sized proteins. This ensures optimal access to the respective target protein, also in crowded environment or 3D samples.
In comparison to conventional secondary reagents the ab325298 nanobody has the following advantages:
- Small probes for enhanced staining speed and improved tissue penetration and higher resolution.
- Monoclonal tools with defined species and isotype specificity.
- Recombinant production for reliable quality, scalability and reproducibility.
- Monovalent binding mode – Pre-incubation without clustering primary antibodies and therefore one-step indirect immunofluorescence possible.
- Enhanced binding allows one-step multi-colour staining and imaging even with primary antibodies from identical species and isotype (Multiplexing).
- Suitable for post-fixation and enhanced performance in demanding application (e.g., expansion microscopy, DNA-PAINT, dSTORM, etc.).
