Biological function summary
Wwp2 plays a critical role in cellular processes like growth division and differentiation by regulating the stability of key signaling proteins. It functions as part of protein complexes that include WW domain-containing proteins which enables it to interact with multiple substrates. Through its ubiquitination activity Wwp2 influences pathways involved in the maintenance of cellular homeostasis and response to stimuli.
Pathways
Wwp2 has significant involvement in the TGF-beta signaling pathway and the Wnt signaling pathway. In the TGF-beta pathway Wwp2 mediates the degradation of SMAD2 and SMAD7 which are central to the regulation of this signaling cascade. Wwp2 also interacts with proteins like SMURF2 and RNF11 within these pathways promoting fine-tuned control of signal transduction ultimately influencing development and differentiation processes.

E3 ubiquitin-protein ligase which accepts ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then directly transfers the ubiquitin to targeted substrates. Polyubiquitinates POU5F1 by 'Lys-63'-linked conjugation and promotes it to proteasomal degradation; in embryonic stem cells (ESCs) the ubiquitination is proposed to regulate POU5F1 protein level. Ubiquitinates EGR2 and promotes it to proteasomal degradation; in T-cells the ubiquitination inhibits activation-induced cell death. Ubiquitinates SLC11A2; the ubiquitination is enhanced by presence of NDFIP1 and NDFIP2. Ubiquitinates RPB1 and promotes it to proteasomal degradation.
