Sumo 2 also known as SUMO-2 or SMT3A is a small ubiquitin-like modifier protein with a molecular mass of approximately 11 kDa. It belongs to the SUMO protein family which includes SUMO-1 SUMO-3 and others. Sumo 2 is widely expressed in the nucleus and cytoplasm of eukaryotic cells. It is involved in the post-translational modification process known as SUMOylation where it covalently attaches to specific lysine residues on substrate proteins influencing their activity localization and stability.

Polymeric SUMO2 chains are also susceptible to polyubiquitination which functions as a signal for proteasomal degradation of modified proteins (PubMed : 18408734, PubMed : 18538659, PubMed : 21965678, PubMed : 9556629). Plays a role in the regulation of sumoylation status of SETX (PubMed : 24105744).
