Biological function summary
A phosphatase that is involved in important tasks within the mitochondria PTPMT1 contributes significantly to the regulation of mitochondrial metabolism. It removes phosphate groups from target substrates which is essential for the biosynthesis of cardiolipin an important lipid in mitochondrial membrane composition. Although not formally part of a large complex it interacts closely with other mitochondrial enzymes to maintain lipid homeostasis.
Pathways
Lipid metabolism and insulin signaling both involve PTPMT1. It mediates its actions through the biosynthesis of phosphatidylinositol derivatives that influence the insulin signaling pathway. It has a connection with the PI3K/Akt signaling cascade affecting the actions of proteins such as Akt and PDK1 which play integral roles in cellular energy balance and glucose homeostasis.

Lipid phosphatase which dephosphorylates phosphatidylglycerophosphate (PGP) to phosphatidylglycerol (PG) (By similarity). PGP is an essential intermediate in the biosynthetic pathway of cardiolipin, a mitochondrial-specific phospholipid regulating the membrane integrity and activities of the organelle (PubMed : 39279645). Has also been shown to display phosphatase activity toward phosphoprotein substrates, specifically mediates dephosphorylation of mitochondrial proteins, thereby playing an essential role in ATP production (By similarity). Has probably a preference for proteins phosphorylated on Ser and/or Thr residues compared to proteins phosphorylated on Tyr residues (By similarity). Probably involved in regulation of insulin secretion in pancreatic beta cells (By similarity). Prevents intrinsic apoptosis, by regulating mitochondrial membrane integrity (PubMed : 24709986, PubMed : 39279645).
