      Procalcitonin (PCT) is a 116 amino acid residue peptide with molecular weight of about 13 kDa. PCT itself has no known hormonal activity. PCT belongs to a group of related proteins including calcitonin gene-related peptides I and II, amylin, adrenomodulin and calcitonin (CAPA peptide family). PCT, like other peptides of CAPA family, appears from the common precursor pre-procalcitonin consisting of 141 amino acids by removal of 25 amino acids from the N-terminus. PCT undergoes successive cleavages to form three molecules: N-terminal fragment (55 a.a.), calcitonin (32 a.a.) and katacalcin (21 a.a.). Under normal metabolic conditions, PCT is only present in the C cells of the thyroid gland. In bacterial infection and sepsis, however, intact PCT is found in the blood and, more importantly, its level is related to the severity of bacterial sepsis. Today, PCT is considered to be one of the earliest and most specific markers of sepsis.

    I have used the rabbit polyclonal to Procalcitonin antibody for IHC and it worked well. I want to submit my manuscript but the journal requires it be crucial to have WB information - either in the species that I am studying or if not feaasible at east in a species like the rat. They suggested to contact you and hear if you have done such control for this antibody? The journal wants to have some kind of control for the antibody such as WB. Hope you can help me with this!

    All the information we have about an antibody we have you usually find on our datasheet. This includes tested species and application. According to the datasheet, WB hasn't been tested, and rat is only predicted to work but not tested. Therefore, we cannot send you any extra information.

    However, I have attached an article in which is described what a good antibody is made of and you will see that all of the demands match this antibody.

    To solve this problem in WB, I would suggest running a positive control in the form of the recombinant protein along your samples. If the signal is positive you can clearly show that the antibody detects the target. In addition, you could also use it instead of a blocking peptide in IHC along a isotype control to see if the specific staining will vanish. I have attached the protocol for this matter.
