The PIM3 protein is a member of the proviral integration site for moloney murine leukemia virus (PIM) serine/threonine kinase family. It is also known as PIM-3 kinase. This protein has a mass of approximately 37 kDa. Expression of PIM3 is found in various tissues including liver pancreas kidney and some cancer cell lines. Scientists recognize its role in phosphorylating target proteins which affects various cellular processes like cell cycle and apoptosis.
Biological function summary

PIM3 functions by regulating cell survival and proliferation. It does not form part of any large protein complex but operates through phosphorylation of substrates. This kinase modulates signaling pathways important for mitogenic stimuli and cellular responses to various stress factors. Tissues with abnormal PIM3 levels may experience altered cell growth and survival which can affect normal physiological processes.

Pathways

PIM3 interacts with cellular signaling networks. It is notably involved in the PI3K/AKT signaling pathway affecting cell growth and metabolism. Additionally PIM3 participates in mTOR signaling linking it to protein synthesis and cellular energy balance. It shows functional relation with proteins such as AKT and mTOR suggesting its integration in these pivotal regulatory circuits.

Excessive PIM3 expression associates with certain types of cancer such as pancreatic cancer and hepatocellular carcinoma. It contributes to oncogenic processes by promoting uncontrolled cell growth and survival. In cancer contexts PIM3 interacts with oncogenes like c-MYC which may exacerbate tumorous characteristics. Targeting PIM3 pathways offers potential therapeutic strategies for related malignancies.

Proto-oncogene with serine/threonine kinase activity that can prevent apoptosis, promote cell survival and protein translation. May contribute to tumorigenesis through : the delivery of survival signaling through phosphorylation of BAD which induces release of the anti-apoptotic protein Bcl-X(L), the regulation of cell cycle progression, protein synthesis and by regulation of MYC transcriptional activity. Additionally to this role on tumorigenesis, can also negatively regulate insulin secretion by inhibiting the activation of MAPK1/3 (ERK1/2), through SOCS6. Involved also in the control of energy metabolism and regulation of AMPK activity in modulating MYC and PPARGC1A protein levels and cell growth.
