GSKIP also known as GSK3β-interacting protein plays a role in the signaling pathways by acting as a scaffold for protein interactions. This protein has an approximate mass of 17 kDa. GSKIP is expressed in various tissues including the brain and heart suggesting its broad functional significance in different physiological contexts. Its ability to interact with Glycogen Synthase Kinase 3 (GSK3) and other proteins highlights its involvement in important cellular processes.
Biological function summary

The protein functions as a regulatory adapter anchoring key kinases to their target proteins. It interacts with GSK3β forming a stable complex that influences several signaling pathways. By modulating these interactions GSKIP plays a role in controlling cell growth differentiation and apoptosis. Its capacity to form complexes with other proteins enhances its functional versatility within the cell impacting numerous biological processes.

Pathways

GSKIP exerts a notable influence in the Wnt/β-catenin and Hedgehog signaling pathways. It interacts with GSK3β which is a critical component of these pathways regulating cellular processes like proliferation and development. By affecting the stability and activity of β-catenin GSKIP indirectly influences gene expression. Additionally its involvement in these pathways links it to other related proteins like Axin which help modulate signal transduction.

GSKIP has connections to cancer and neurodegenerative diseases partly due to its role in regulating GSK3β activity. Alterations in the pathways involving GSKIP can impact disease progression and response to treatment. In cancer dysregulation of the Wnt pathway through GSKIP interactions may lead to abnormal cell proliferation. In neurodegenerative diseases its interaction with GSK3β relates to the phosphorylation of tau protein a significant aspect of Alzheimer's disease pathology.

常规信息

功能

A-kinase anchoring protein for GSK3B and PKA that regulates or facilitates their kinase activity towards their targets (PubMed : 16981698, PubMed : 25920809, PubMed : 27484798). The ternary complex enhances Wnt-induced signaling by facilitating the GSK3B- and PKA-induced phosphorylation of beta-catenin leading to beta-catenin degradation and stabilization respectively (PubMed : 16981698, PubMed : 27484798). Upon cAMP activation, the ternary complex contributes to neuroprotection against oxidative stress-induced apoptosis by facilitating the PKA-induced phosphorylation of DML1 and PKA-induced inactivation of GSK3B (PubMed : 25920809). During neurite outgrowth promotes neuron proliferation; while increases beta-catenin-induced transcriptional activity through GSK3B kinase activity inhibition, reduces N-cadherin level to promote cell cycle progression (PubMed : 19830702).
