Glycogenin 1 also known as glycogen protein or glycogenin protein plays an essential role in the biosynthesis of glycogen. Glycogenin 1 initiates the primer strand for glycogen synthesis by self-glucosylation which is the first step in building a glycogen molecule. Glycogenin 1 typically has a molecular mass of approximately 37 kDa. It expresses primarily in tissues with high energy demands such as liver and skeletal muscle. Glycogen synthase 2 on the other hand is an enzyme involved in elongating the glycogen chain once initiated by Glycogenin 1 working closely together in glycogen metabolism.

Glycogenin 1 and glycogen synthase 2 play significant roles in glycogen metabolism. Glycogenin 1 starts glycogen synthesis by adding glucose units to itself forming the core structure of glycogen granules.
