EXOSC8 also known as exosome component 8 forms part of the core of the exosome complex. This protein has a molecular weight of approximately 27 kDa. It is expressed in various tissues with enriched expression in bone marrow. EXOSC8 functions as an important structural component of the exosome complex aiding in the degradation and processing of RNA molecules.
Biological function summary

EXOSC8 plays a central role in RNA metabolism as part of the exosome complex which is essential for RNA processing and turnover. It assists in the degradation of both precursor mRNA and aberrantly processed RNA. The exosome complex typically includes nine core components with EXOSC8 acting alongside these to help maintain RNA stability and facilitate gene expression regulation.

Pathways

EXOSC8 is involved in RNA processing pathways essential for cellular function and survival. These pathways such as the RNA degradation pathway and mRNA surveillance pathway ensure the quality and regulation of RNA species. EXOSC8 interacts closely with other exosome components like EXOSC3 and EXOSC9 enhancing RNA degradation and contributing to cellular homeostasis.

Mutations in EXOSC8 link to neurological conditions notably pontocerebellar hypoplasia type 1B. This rare genetic disorder affects brain development and function leading to severe developmental delays. EXOSC8 mutations can disrupt normal exosome function impacting proteins like EXOSC3 which may worsen disease severity. Another disorder associated with EXOSC8 is autosomal recessive cerebellar ataxia where defective RNA processing contributes to the disease pathology.

常规信息

功能

Non-catalytic component of the RNA exosome complex which has 3'->5' exoribonuclease activity and participates in a multitude of cellular RNA processing and degradation events. In the nucleus, the RNA exosome complex is involved in proper maturation of stable RNA species such as rRNA, snRNA and snoRNA, in the elimination of RNA processing by-products and non-coding 'pervasive' transcripts, such as antisense RNA species and promoter-upstream transcripts (PROMPTs), and of mRNAs with processing defects, thereby limiting or excluding their export to the cytoplasm. The RNA exosome may be involved in Ig class switch recombination (CSR) and/or Ig variable region somatic hypermutation (SHM) by targeting AICDA deamination activity to transcribed dsDNA substrates. In the cytoplasm, the RNA exosome complex is involved in general mRNA turnover and specifically degrades inherently unstable mRNAs containing AU-rich elements (AREs) within their 3' untranslated regions, and in RNA surveillance pathways, preventing translation of aberrant mRNAs. It seems to be involved in degradation of histone mRNA. The catalytic inactive RNA exosome core complex of 9 subunits (Exo-9) is proposed to play a pivotal role in the binding and presentation of RNA for ribonucleolysis, and to serve as a scaffold for the association with catalytic subunits and accessory proteins or complexes. EXOSC8 binds to ARE-containing RNAs.
