The EMG1 protein also known as Bystin-like protein or N1-methyladenosine methyltransferase non-catalytic subunit EMG1 functions as a ribosome biogenesis factor. It weighs approximately 28 kDa. EMG1 localizes mostly in the nucleolus of cells where ribosomal assembly occurs. Its expression occurs in various tissues with significant levels observed in actively dividing cells such as those in proliferative tissues.
Biological function summary

EMG1 serves as an integral component of the small subunit processome a large ribonucleoprotein complex. Within this complex EMG1 participates in properly folding and processing precursor rRNA (pre-rRNA). This step is essential for the maturation and functionality of the small ribosomal subunit which is necessary for protein synthesis.

Pathways

The involvement of EMG1 in ribosome biogenesis places it in the pre-rRNA processing pathway. EMG1 interacts with proteins such as UTP18 and has connections to the snoRNA-guided modification system. These interactions facilitate the endonucleolytic cleavage of pre-rRNA ensuring accurate and efficient production of ribosomal subunits.

Mutations or dysregulation of EMG1 affect ribosome biogenesis possibly leading to conditions like Bowen-Conradi syndrome. This syndrome results from a specific mutation in the EMG1 gene and is marked by developmental abnormalities. Moreover disruptions in EMG1 link it indirectly to ribosomopathies disorders characterized by impaired ribosomal function.

S-adenosyl-L-methionine-dependent pseudouridine N(1)-methyltransferase that methylates pseudouridine at position 1248 (Psi1248) in 18S rRNA. Involved the biosynthesis of the hypermodified N1-methyl-N3-(3-amino-3-carboxypropyl) pseudouridine (m1acp3-Psi) conserved in eukaryotic 18S rRNA. Is not able to methylate uridine at this position (PubMed : 20047967). Has also an essential role in 40S ribosomal subunit biogenesis independent on its methyltransferase activity, facilitating the incorporation of ribosomal protein S19 during the formation of pre-ribosomes (By similarity). Part of the small subunit (SSU) processome, first precursor of the small eukaryotic ribosomal subunit. During the assembly of the SSU processome in the nucleolus, many ribosome biogenesis factors, an RNA chaperone and ribosomal proteins associate with the nascent pre-rRNA and work in concert to generate RNA folding, modifications, rearrangements and cleavage as well as targeted degradation of pre-ribosomal RNA by the RNA exosome (PubMed : 34516797).
