The agouti signaling protein (ASIP) also known as agouti protein or agouti switch protein functions mechanically as an antagonist to the melanocortin 1 receptor (MC1R). This interaction influences pigmentation processes by modulating eumelanin and pheomelanin production. ASIP possesses a mass of approximately 14 kDa. It is expressed in several tissues such as skin adipose tissue and neural tissues highlighting its varied roles in different biological contexts.
Biological function summary

ASIP plays an important role in pigmentation regulation and energy homeostasis. It prevents MC1R from binding its ligand alpha-melanocyte-stimulating hormone (α-MSH) leading to lightening of skin and fur coloration by shifting melanin production from eumelanin to pheomelanin. Although ASIP is not part of a larger protein complex its interaction with the MC1R receptor greatly influences pigmentation phenotypes.
