      - First try to dissolve a small amount of peptide in either water or buffer. The more charged residues on a peptide, the more soluble it is in aqueous solutions.
      - If the peptide doesn’t dissolve try an organic solvent e.g. DMSO, then dilute using water or buffer.
      - Consider that any solvent used must be compatible with your assay. If a peptide does not dissolve and you need to recover it, lyophilise to remove the solvent.
      - Gentle warming and sonication can effectively aid peptide solubilisation. If the solution is cloudy or has gelled the peptide may be in suspension rather than solubilised.
      - Peptides containing cysteine are easily oxidised, so should be prepared in solution just prior to use.

    We used this peptide as a positive control for an anti-netrin 1 peptide antibody in an ELISA assay. The peptide worked very well. We also found that the peptide as a potent chemorepellant in our cell system. We plan to use this peptide as a positive control for our Western Blots as well.

    I spoke to my colleague Jill who is in charge to source new proteins, and even she agrees that we should have this protein in our catalog, I am afraid this will take quite a while ( more than two month at least).

    Therefore, I am afraid we cannot help you with a full length protein you can use as a standard right now when you need it.

    We would need recombinant Sema 3c for making the standard curve. Do you think that Semaphorin 3c peptide (ab 39299) would be suitable to use in the sandwich ELISA to make the standard curve?

    I think the problem with this peptide is that it will probably only be detected by the polyclonal. I cannot tell you if the monoclonal antibody can bind this peptide as the epitope wasn't mapped.

    However, I have asked our protein expert if it is possible to get our hands on the full length proteinand will let you know if I know anything more.
