Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. The carrier-free buffer and high concentration allow for increased conjugation efficiency.
This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

xCT has a predicted molecular weight of 55 kDa; however it has a high number of hydrophobic residues which may affect the migration of the protein in SDS-PAGE. Endogenous monomeric xCT is expected to migrate at ~35 kDa and modified-xCT is expected to migrate at ~55 kDa (PMID: 17035536) -

The expression profile observed is consistent with what has been described in the literature ( PMID: 32188872). -
This data was developed using ab300667, the same antibody clone in a different buffer formulation.
Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized A549 (human lung carcinoma epithelial cell) cells labeling xCT with AB300667 at 1/50 dilution (21.18 μg/ml), followed by ab150157 Goat Anti-Rat IgG H&L (Alexa Fluo®488) antibody at 1/1000 dilution (2 μg/mL) (Green). Confocal image showing membranous and cytoplasmic staining in A549 cell line. Low expression: colo 205 did not show any stainine (PMID: 31000598). Anti-beta Tubulin rabbit monoclonal antibody (ab179513) was used to counterstain tubulin at 1/200 dilution (10 μg/mL) (Red). The Nuclear counterstain was DAPI (Blue).
The negative controls are as follows:
-ve control 1: ab300667 at 1/50 dilution, followed by secondary antibody name (ab150080) secondary at 1/500 dilution (4 μg/ml).
-ve control 2: Primary (ab179513) at 1/200 dilution, followed by secondary antibody name (ab150157) secondary at 1/1000 dilution (2 μg/ml). -
This data was developed using ab300667, the same antibody clone in a different buffer formulation.
xCT was immunoprecipitated from 0.35 mg HepG2 (human hepatocellular carcinoma epithelial cell) whole cell lysate with ab300667 at 1/30 dilution (2 µg in 0.35 mg lysates). Western blot was performed on the immunoprecipitate using ab300667 at 1/1000 dilution. Goat Anti-Rat IgG (H+L), HRP) (ab205720) was used at 1/10000 dilution.
