UTP18 was immunoprecipitated from HeLa (human epithelial cell line from cervix adenocarcinoma) whole cell lysate (1 mg for IP, 20% of IP loaded) with ab241578 at 3 μg/mg lysate. Western blot was performed from the immunoprecipitate using ab241578 at 0.1 μg/ml.

Lane 1 : ab241578 IP in HeLa whole cell lysate.

Lane 2 : Control IgG IP in HeLa whole cell lysate.

Detection : Chemiluminescence with exposure time of 1 second.

Lysates prepared using NETN lysis buffer.

UTP18 also known as U3 small nucleolar RNA-associated protein 18 plays a mechanical role in the assembly and maturation of small subunit (SSU) ribosomal RNA. UTP18 with an approximate mass of 35 kDa functions as part of the UTP-B module of the SSU processome. It is expressed primarily in the nucleolus of eukaryotic cells where it facilitates rRNA synthesis and processing an essential step in ribosome biogenesis.
Biological function summary

UTP18 contributes to the early processing stages of the 18S rRNA and is integral to the formation of the SSU processome. This protein is part of a larger complex known as the small subunit (SSU) processome which contains several U3 small nucleolar RNAs and numerous protein subunits. Its activity supports the assembly of ribosomal subunits an essential function for cellular protein synthesis and growth.

Pathways

UTP18 participates in ribosome biogenesis and nucleotide metabolism in eukaryotic cells. It associates with various proteins such as UTP14 and others within the SSU processome to ensure proper rRNA processing. UTP18 action affects the ribosome biogenesis pathway directly impacting translation and cellular metabolism.

Mutations or dysregulation of UTP18 have links to ribosomopathies which involve impaired ribosome function. Disorders such as Diamond-Blackfan anemia characterized by defective erythropoiesis may involve issues with UTP18 function. Its interactions with other SSU processome components suggest that any disruptions in this protein complex might contribute to disease pathology affecting ribosomal assembly and cellular protein synthesis.

Part of the small subunit (SSU) processome, first precursor of the small eukaryotic ribosomal subunit. During the assembly of the SSU processome in the nucleolus, many ribosome biogenesis factors, an RNA chaperone and ribosomal proteins associate with the nascent pre-rRNA and work in concert to generate RNA folding, modifications, rearrangements and cleavage as well as targeted degradation of pre-ribosomal RNA by the RNA exosome.
