UBR3 also known as UBR domain-containing protein 3 functions as an important E3 ubiquitin ligase within the N-end rule pathway. It has a molecular mass of approximately 200 kDa. UBR3 is widely expressed with significant expression in the brain heart and skeletal muscle tissues. The protein plays an important role in recognizing and tagging proteins that display destabilizing N-terminal residues for proteasomal degradation a process essential for maintaining protein homeostasis.
Biological function summary

This protein performs essential roles in protein quality control and turnover. UBR3 is part of the ubiquitin-proteasome system and forms part of larger ubiquitin ligase complexes. This system governs the degradation of most intracellular proteins regulating various cellular processes like cell cycle progression and apoptosis. It ensures that damaged or misfolded proteins which could disrupt cellular functions are promptly identified and degraded.

Pathways

The ubiquitin proteasome pathway is one of the most important biological frameworks involving UBR3. This pathway pivotal for regulating protein concentrations and controlling cellular functions connects UBR3 with other proteins such as UBR1 and UBR2 which together function in substrate recognition and ubiquitination. Additionally UBR3 is involved in the N-end rule pathway further tying it to numerous cellular activities related to protein synthesis modification and degradation.

Irregularities in UBR3 function could contribute to neurological conditions and muscular degenerative disorders. Altered activity or expression of UBR3 might lead to Neurological disorders such as certain types of ataxias possibly due to disrupted protein homeostasis in neurons. Connections with proteins like p53 in pathways regulated by ubiquitin ligases also implicate UBR3 in cancer where it could affect cell cycle control and apoptosis thereby influencing tumor growth and progression.

Does not bind to proteins bearing specific N-terminal residues that are destabilizing according to the N-end rule, leading to their ubiquitination and subsequent degradation (By similarity). May play a role in Shh signaling by mediating the ubiquitination of Kif7 (By similarity). May be important for MYH9 function in certain tissues, possibly by regulating the ubiquitination of MYH9 and consequently affecting its interaction with MYO7A (PubMed : 27331610).
