    IHC image of Tubby staining in human cerebral cortex formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab46943, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

I just got the information from my lab colleagues.

They confirmed the mouse strain used is CD-1 and the animals used are surplus animals over 8 weeks old.

Hello, I bought this anti-tub (ab 46943) and I can not detect the TUB protein in a sample of normal mouse brain (but in testis, yes !) I am sorry you are experiencing difficulties with this antibody.

The lysates were prepared using RIPA buffer and phosphatase inhibitors. The gel was run under reducing conditions, and the blocking was perform in 5%BSA/0.1%TBST for one hour with gentle rocking. Antibodies are incubated overnight at 4 degrees C in blocking buffer (5%BSA/0.1%TBST). The membrane was washed with 0.1% TBST 3x10minutes at RT.

The procedure was a standard WB protocol as described in our online protocol:
