TRPM7 and TRPM6 are members of the transient receptor potential melastatin subfamily often referred to as TRP-PLIK or CHAK1 for TRPM7. TRPM7 has a molecular mass of approximately 165 kDa while TRPM6 is roughly 230 kDa. These proteins function as ion channels with kinase activity. They regulate the homeostasis of divalent cations like magnesium and calcium by permitting their entry into cells. TRPM7 is expressed in various tissues including the heart liver and brain while TRPM6 is mostly found in the kidney and intestine.
Biological function summary

TRPM7 and TRPM6 play essential roles in cellular magnesium absorption and homeostasis. They form homo- or heteromeric channel complexes with the combination of TRPM6 and TRPM7 enhancing channel function. These proteins influence critical cellular processes such as proliferation apoptosis and migration by modulating cellular magnesium levels. TRPM7's distinct kinase domain also impacts intracellular signaling pathways through phosphorylation of specific substrates regulating response to cellular stress and differentiation.

Pathways

TRPM7 and TRPM6 modulate cell signaling pathways linked to magnesium-sensitive enzymes and proteins like those involved in the PI3K/Akt signaling pathway. This pathway is integral for cell growth survival and metabolism. TRPM7 interacts with various signaling proteins including PLC and MAPK influencing cellular responses to external stimuli. TRPM6 often partners with TRPM7 to fine-tune these cellular pathways maintaining appropriate ionic balances necessary for cellular functions.

TRPM7 and TRPM6 are linked to magnesium homeostasis-related conditions such as hypomagnesemia and hypertension. Variants in TRPM6 associate with hypomagnesemia with secondary hypocalcemia a genetic disorder affecting magnesium uptake in the intestine. TRPM7 is connected to neurodegenerative diseases given its role in neuronal survival and response to oxidative stress. The interaction of TRPM7 with proteins involved in neurodegenerative pathways like amyloid precursor protein suggests potential involvement in Alzheimer’s disease pathogenesis.

靶点信息

Bifunctional protein that combines an ion channel with an intrinsic kinase domain, enabling it to modulate cellular functions either by conducting ions through the pore or by phosphorylating downstream proteins via its kinase domain. The channel is highly permeable to divalent cations, specifically calcium (Ca2+), magnesium (Mg2+) and zinc (Zn2+) and mediates their influx (PubMed : 11385574, PubMed : 12887921, PubMed : 15485879, PubMed : 24316671, PubMed : 35561741, PubMed : 36027648). Controls a wide range of biological processes such as Ca2(+), Mg(2+) and Zn(2+) homeostasis, vesicular Zn(2+) release channel and intracellular Ca(2+) signaling, embryonic development, immune responses, cell motility, proliferation and differentiation (By similarity). The C-terminal alpha-kinase domain autophosphorylates cytoplasmic residues of TRPM7 (PubMed : 18365021). In vivo, TRPM7 phosphorylates SMAD2, suggesting that TRPM7 kinase may play a role in activating SMAD signaling pathways. In vitro, TRPM7 kinase phosphorylates ANXA1 (annexin A1), myosin II isoforms and a variety of proteins with diverse cellular functions (PubMed : 15485879, PubMed : 18394644).. TRPM7 channel, cleaved form. The cleaved channel exhibits substantially higher current and potentiates Fas receptor signaling.. TRPM7 kinase, cleaved form. The C-terminal kinase domain can be cleaved from the channel segment in a cell-type-specific fashion. In immune cells, the TRPM7 kinase domain is clipped from the channel domain by caspases in response to Fas-receptor stimulation. The cleaved kinase fragments can translocate to the nucleus, and bind chromatin-remodeling complex proteins in a Zn(2+)-dependent manner to ultimately phosphorylate specific Ser/Thr residues of histones known to be functionally important for cell differentiation and embryonic development.
