TRH-R also known as Thyrotropin-Releasing Hormone Receptor is a G-protein-coupled receptor specifically binding the tripeptide hormone TRH. This receptor has a molecular weight of approximately 70 kDa. It is express in high levels in the anterior pituitary gland as well as the central nervous system and various peripheral tissues. TRH-R plays a mechanical role in mediating the effects of TRH which include stimulating the release of thyroid-stimulating hormone (TSH) and prolactin from the anterior pituitary.
Biological function summary

The TRH-R is involved in important physiological processes. The binding of TRH to TRH-R initiates a cascade of intracellular events leading to the activation of phospholipase C and the subsequent production of inositol triphosphate and diacylglycerol. This signaling pathway elevates intracellular calcium levels and ultimately modulates the secretion of hormones like TSH. TRH-R does not form part of a larger complex but relies on interaction with heterotrimeric G proteins to exert its effects.

Pathways

TRH-R is significantly involved in the hypothalamic-pituitary-thyroid axis. Upon activation by TRH TRH-R activates signaling pathways that regulate thyroid hormone levels playing a role in metabolism. It also interacts with other receptors such as somatostatin receptors which can modulate the inhibitory effects on hormone secretion processes.

The dysregulation of TRH-R function has links with conditions like hypothyroidism and hyperprolactinemia. In these disorders the altered signaling of TRH-R influences the levels of important hormones such as TSH and prolactin. TRH-R connects with thyrotropin and prolactin-related proteins which serves as both diagnostic markers and potential therapeutic targets in these diseases.
