    Transferrins are iron binding transport proteins which can bind two Fe(3+) ions in association with the binding of an anion, usually bicarbonate. It is responsible for the transport of iron from sites of absorption and heme degradation to those of storage and utilization. Serum transferrin may also have a further role in stimulating cell proliferation.

      Lane 1 : Human Transferrin – 8 µl (reducing conditions) at 5 µg
      Lane 2 : HumanTransferrin – 5 µl (reducing conditions) at 3 µg
      Lane 3 : Human Transferrin – 3 µl (reducing conditions) at 1 µg
      Lane 4 : Human Transferrin – 3 µl (non reducing conditions) at 1 µg
      Lane 5 : Human Transferrin – 5 µl (non reducing conditions) at 3 µg

      The sample (commercial human transferrin) (1µg/ml in 1x PBS) was mixed with 4x loading buffer (non reducing) or with Laemmli buffer (reducing) and warmed for 3 minutes.

      Volume of sample:5; 3; 1 µl of sample + 3; 2; 2 µl of buffer

      SDS Page: 12% AA-gel, U=180V, 90 minutes, volume of loading sample:8; 5;3 µl

      Marker – 3 µl

      WB: Nitrocelulose membrane (8x5.5 cm), I=35mA, 90minutes

      Detection:

      Blocking solution: 1xPBS/5% BSA, 4°C, overnight

      Primary Ab: membrane was incubated in 5 ml DMEM/10%FTS + 10 µl HTF-14 (primary Ab) (60 minutes, room temperature)

      Washing solution: 1x PBS/0.1% Tween

      Secondary Ab: IRDye® 800CW Goat anti-mouse IgG1-specific, dilution 1/5000, diluting solution – 5% BSA/1x PBS, incubation for 60 minutes, low temperature.

      Detection: Li-COR odyssey

      No reducing agent should be added to the sample buffer. The sample can be denatured and processed by common Western blotting protocols, but the disulfide bonds must not be reduced.

    Question

    We wish to create a lateral flow assay, I am shopping for antibodies against human transferrin. Please help me on following questions.  1) Antibody pairing information to generate lateral flow test for detecting human transferrin  2) If such pairing information is available, we would like to know the performance of such pair in lateral flow test, i.e. the sensitivity and specificity   (crossreactivity with transferrin from other species).  3) Positive control information to validate the antibody performance in lateral flow test.

    Lateral flow is not a tested application, however it is very much like sandwich ELISA so any ab pair used in sELISA should work in LF. We do offer a matched antibody for use with ab769 this is ab9538. Gold conjugation is generally used in LF. We do not carry gold conjugated anti-Transferrin, so the customer will need to conjugate one ab for use as the colored marker. 1) Ab9538 is tested and guaranteed in sELISA. For use in LT we recommend using this ab9583 as the test line and ab769 as colored particle. We do not have a gold conjugation kit however these may be found at online. Control line will be Mouse IgG1 monoclonal antibody [NCM1] - Isotype control (ab27479). You will need a detection system which will be able to read the gold particles, a flatbed scanner will likely work. 2) We do not test in LT. Ab769 is known to Reacts with: Rabbit, Human, Pig and not with Sheep, Horse, Cow, Dog or Carp. AB9538 is known to react with human and Marmoset; other species have not been tested. 3) Transferrin protein (Active) ab83560 has been tested with and is the positive control for sELISA with this pair.

    Since all three antibodies have IgG (or a IgG subclass) isotypes, the molecular weight of each is approximately 160kDa (23-25kDa for both light chains, and 53kDa for both heavy chains).

    Thank you for the requested information. I have re-examined our record and I was not able to find this information in our past correspondences with you. In regards to the purchase order number "192677", this was an order for ab7355 (Acinus antibody) placed by Patricia from the Inserm. Could you please check this information again and provide the correct PO# for ab769? Our policy at Abcam is that if an antibody does not work as specified on the datasheet, we will offer a replacement or reimbursement if the product has been purchased within 90 days.
