    Defects in TPO are the cause of congenital hypothyroidism due to dyshormonogenesis type 2A (CHDH2A) [MIM:274500]; also called genetic defect in thyroid hormonogenesis 2A or thyroid hormone organification defect II. CHDH2A is due to defective conversion of accumulated iodide to organically bound iodine. The iodide organification defect can be partial or complete.

      Immunohistochemical staining of paraffin embedded human thyroid with purified ab133322 at a working dilution of 1 in 1000. The secondary antibody used is a HRP polymer for rabbit IgG. The sample is counter-stained with hematoxylin. Antigen retrieval was perfomed using Tris-EDTA buffer, pH 9.0. PBS was used instead of the primary antibody as the negative control, and is shown in the inset.

    Question

    Our lab has been trying to detect TPO in bronchial epithelial cells. We used TPO antibody -- ab133322 (1:500) and used TPO lysate from another company (15 ul) and human thyroid tissue (15 ul) as positive controls. Unfortunately, no band showed up with TPO lysate, and there is a band at ~ 25 kDa with human thyroid tissue (photo attaced). The expected molecular weight of TPO is ~ 100 kDa. We tried a few times and results were the same. So I would need your suggestion on this regard. One more thing, could you provide more detail about the immunogen used for this antibody (e.g. epitope, amino acid sequence) ? Thanks very much !
