补充信息

THRAP5 also known as TRIP13 and thyroid hormone receptor-associated protein 5 is a protein with a molecular mass of approximately 36 kDa. This protein usually operates in the nucleus as a part of transcriptional regulation processes. It is strongly expressed in various tissues including the liver kidney and brain indicating its widespread presence in the body. Mechanically THRAP5 interacts with nuclear hormone receptors and acts as a coactivator which means that it helps facilitate the transcription of specific genes.
Biological function summary

THRAP5 functions as part of a larger complex called the Mediator complex which is essential for the regulation of gene expression. This complex serves as a bridge linking transcriptional activators and the RNA polymerase II machinery thereby influencing the transcription of genes downstream. THRAP5 is involved in various regulatory roles that include assisting in the activation of nuclear receptors. Its involvement in the complex structure of Mediator indicates its role in controlling gene expression.

Pathways

THRAP5 involves itself in the regulation of transcriptional processes and may influence the ERK/MAPK signaling pathway. This pathway is important for cell growth proliferation and differentiation. The protein interacts with transcription factors and other proteins like ERK1/2 to modulate these pathways. Its role in facilitating communication between hormone receptors and transcriptional machinery positions it as an important regulatory hub in cellular signaling.

THRAP5 interacts with various proteins involved in cancer and metabolic disorders. Studies have linked its aberrant expression to certain cancers including breast cancer where it might play a role in tumor progression. Additionally metabolic conditions like obesity have shown connections to THRAP5 potentially through interactions with PPARs (peroxisome proliferator-activated receptors) which are important for metabolic regulation. Understanding the contribution of THRAP5 in these diseases could offer insights into novel therapeutic targets.

靶点信息

Component of the Mediator complex, a coactivator involved in the regulated transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II and the general transcription factors.
