As we discussed, the molecular weight of the TGF beta-1 protein is about 45 kDa, which is the predicted band size for this antibody. He estudiado el protocolo enviado, y hay una serie de sugerencias que me gustaría hacer para intentar mejorar los resultados del anticuerpo.

Es posible que no se observen bandas al incubar con el anticuerpo debido a un exceso en las condiciones de bloqueo. Mi recomendación es usar 5% BSA para bloquear en lugar de leche. Incubar durante 1 hora a TA con este agente. Además, para diluir los anticuerpos (primario y secundario) yo os sugeriría usar el mismo agente que para bloquear, es decir BSA, pero a una concentración menor, como 3%.
Aumentar la concentración de primario, a 1/100.
Solo por confirmar, ¿es el secundario es especifico contra rabbit?

From the blot you have shared with me, it does indeed look like you are detecting the protein as expected, especially in the human thrombocyte sample. From having reviewed the protocol used to obtain this blot there are a few suggestions I would make which may improve the non-specificity observed. As you have tried BSA and the ECL blocking agent with non-specificity observed, I would suggest also trying 5% milk blocking. This can sometimes have a surprisingly marked effect.

As you have mentioned, trying a higher dilution of the primary antibody may also help and increasing the eletrophoresis time will allow you to identify the bands of interest more clearly.
