Streptolysin is a toxin produced by certain strains of Streptococcus bacteria including the well-known Streptococcus pyogenes. This protein acts as a hemolysin creating pores in the host cell membranes leading to cell lysis. Streptolysin comes in two main forms Streptolysin O (SLO) and Streptolysin S (SLS). SLO is sensitive to oxygen and has an approximate mass of 69 to 74 kDa. Unlike SLS which is stable in the presence of oxygen. These toxins are mainly expressed during bacterial infection and play a role in promoting bacterial virulence.
Biological function summary

These hemolysins disrupt cellular integrity by forming large oligomeric pores in host cell membranes. SLO is part of the cholesterol-dependent cytolysin family requiring cholesterol-rich membranes for activity. While it is not part of a permanent complex SLO can form a temporary assembly with other molecules to execute its pore-forming function. This results in significant effects on cells and tissues infected by Streptococcus.

Pathways

Both forms of streptolysin interfere with host immune system pathways and cellular signaling. They can disrupt immune functions allowing bacteria to evade host defenses. Among the important pathways influenced by SLO is the complement pathway which plays an important role in immune response. Streptolysin can indirectly affect proteins like complement component C5a leading to reduced recruitment of immune cells to the infection site.

Streptolysin is closely linked to conditions like pharyngitis and rheumatic fever. These toxins contribute to the severe outcomes associated with these infections. Moreover streptolysin has been associated with damage to cardiac tissue in rheumatic fever possibly interacting with cardiac myosin a protein involved in heart muscle contraction. This connection makes streptolysin an interesting target for research in therapies aimed at mitigating its harmful effects.
