ST3GAL2 also known as ST3 beta-galactoside alpha-23-sialyltransferase 2 is an enzyme encoded by the ST3GAL2 gene. It is involved in the transfer of sialic acid onto galactose-containing oligosaccharides. The protein has a molecular mass of approximately 38 kDa. ST3GAL2 is expressed in a variety of tissues with higher expression reported in the brain and placenta. It localizes mainly to the Golgi apparatus where it participates in the modification of glycoproteins and glycolipids.
Biological function summary

The enzyme ST3GAL2 contributes to the biosynthesis of sialylated glycoconjugates which are important in cellular communication and stability. It plays a role in producing sialyl Lewis X (sLeX) a ligand involved in cell adhesion. ST3GAL2 operates as part of the Golgi glycosylation machinery and influences the functional properties of cell surface molecules. Its activity impacts processes such as immune response and signaling pathways where the proper display of sialylated structures is required for interacting with selectins.

Pathways

ST3GAL2 plays a vital role in the glycosylation and glycan biosynthesis pathways. It impacts the O-glycan biosynthesis pathway where it contributes to the capping of glycan chains with sialic acid. This sialylation process affects the recognition and binding activities of selectins a family of cell adhesion proteins. In particular ST3GAL2 interacts with other enzymes like ST3GAL1 and ST6GAL1 both of which participate in creating complex glycan structures.

ST3GAL2 associates with cancer and neurological disorders. Altered sialylation patterns driven by overexpression or underexpression of ST3GAL2 can influence tumor progression and metastasis. In cancer changes in glycan structures affect interactions with the immune system and modulate cell migration. Additionally ST3GAL2 connects to disorders involving the immune system as modifications in glycan structures can impact the function of immune-related proteins like selectins influencing inflammatory response and immune escape mechanisms in tumor environments.

A beta-galactoside alpha2-3 sialyltransferase primarily involved in terminal sialylation of ganglio and globo series glycolipids (PubMed : 8920913, PubMed : 9266697). Catalyzes the transfer of sialic acid (N-acetyl-neuraminic acid; Neu5Ac) from the nucleotide sugar donor CMP-Neu5Ac onto acceptor Galbeta-(1->3)-GalNAc-terminated glycoconjugates through an alpha2-3 linkage (PubMed : 25916169, PubMed : 8920913, PubMed : 9266697). Sialylates GM1/GM1a, GA1/asialo-GM1 and GD1b gangliosides to form GD1a, GM1b and GT1b, respectively (PubMed : 8920913, PubMed : 9266697). Together with ST3GAL3, primarily responsible for biosynthesis of brain GD1a and GT1b that function as ligands for myelin-associated glycoprotein MAG on axons, regulating MAG expression and axonal myelin stability and regeneration (By similarity). Via GT1b regulates TLR2 signaling in spinal cord microglia in response to nerve injury (By similarity). Responsible for the sialylation of the pluripotent stem cell- and cancer stem cell-associated antigen SSEA3, forming SSEA4 (PubMed : 12716912). Sialylates with low efficiency asialofetuin, presumably onto O-glycosidically linked Galbeta-(1->3)-GalNAc-O-Ser (PubMed : 25916169, PubMed : 9266697).
