      Transcriptional activator required for lipid homeostasis. Regulates transcription of the LDL receptor gene as well as the fatty acid and to a lesser degree the cholesterol synthesis pathway (By similarity). Binds to the sterol regulatory element 1 (SRE-1) (5'-ATCACCCCAC-3'). Has dual sequence specificity binding to both an E-box motif (5'-ATCACGTGA-3') and to SRE-1 (5'-ATCACCCCAC-3').

      Expressed in a wide variety of tissues, most abundant in liver and adrenal gland. In fetal tissues lung and liver shows highest expression. Isoform SREBP-1C predominates in liver, adrenal gland and ovary, whereas isoform SREBP-1A predominates in hepatoma cell lines. Isoform SREBP-1A and isoform SREBP-1C are found in kidney, brain, white fat, and muscle.

      At low cholesterol the SCAP/SREBP complex is recruited into COPII vesicles for export from the ER. In the Golgi complex SREBPs are cleaved sequentially by site-1 and site-2 protease. The first cleavage by site-1 protease occurs within the luminal loop, the second cleavage by site-2 protease occurs within the first transmembrane domain and releases the transcription factor from the Golgi membrane. Apoptosis triggers cleavage by the cysteine proteases caspase-3 and caspase-7.
      Phosphorylated by AMPK, leading to suppress protein processing and nuclear translocation, and repress target gene expression. Phosphorylation at Ser-402 by SIK1 represses activity possibly by inhibiting DNA-binding.

        This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 55 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was blocked for an hour using 5% milk before ab3259 and ab181602 (rabbit anti-GAPDH loading control) were incubated overnight at 4°C at a 5ug/ml concentration and 1/20000 dilution respectively. Antibody binding was detected using Goat anti-Rabbit IgG H&L (IRDye® 680RD) preadsorbed (ab216777) and Goat anti-Mouse IgG H&L (IRDye® 800CW) preadsorbed (ab216772) secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

      Muchas gracias por contactarnos de nuevo.

      He llevado a cabo un BLAST entre las secuencias del inmunógeno usado para generar ab3259 y la secuencia de la proteína en conejo, y el porcentaje de homología es bastante elevado (91%), por lo que es muy probable que el anticuerpo funcione en esta especie.

      El otro anticuerpo, ab135196, nos lo facilita un colaborador, y la secuencia del inmunógeno utilizada es considerada confidencial, por lo que no he podido llevar a cabo la alineación de secuencias. He comparado la secuencia total de las proteínas en humano y en conejo, y no es muy alta (76%), pero este dato no es determinante para predecir la reactividad cruzada, puesto que no sabemos qué porción de la secuencia se usó para producir el anticuerpo.

      En cualquier caso, si te interesa testar los anticuerpos en conejo, puedo mandarte los códigos canjeables por otros productos si participas en el sistema Abreview. Recuerda que antes de comprar los anticuerpos es necesario emitir dichos códigos.

      SREBP ab 3259, lot no: GR81494-1 I had performed western blot on rat liver lysate(whole cell lysate). Lysate was prepared fresh in RIPA buffer with proteolytic inhibitors. 120µg of protein was loaded per well on a 7% SDS-PAGE with 4% stacking. Blocking: 1 hour at 25ºC in 5% milk TBST(0.1%) Primary antibody: 1:400 in 2.5% milk TBST(0.1%) - 16hrs at 4ºC. Secondary antibody: 1:1000 anti mouse IgG in 5% milk TBST(0.1%) Developed using west pico chemiliminescent substrate Results: fig 1. Prominent band almost at the dye front (approx 66Kda).Faint band at 97Kda, prominent only in the 5th lane. Query: The predicted molecular weight of SREBP1 is 127Kda. There is no band of the expected molecular weight. Lanes 1-5: rat liver whole cell lysate The experiment was repeated using nuclear extract. 50 µg of protein was loaded per well on a 10% SDS-PAGE with 4% stacking. Blocking: 1 hour at 25ºC in 5% milk TBST(0.1%) Primary antibody: 1:400 in 2.5% milk TBST(0.1%) - 16hrs at 4ºC. Secondary antibody: 1:1000 anti mouse IgG in 5% milk TBST(0.1%) Developed using west pico chemiliminescent substrate – 12 min exposure. Result: fig 2. Prominent band at 30Kda. Other bands at 34,56 and 97 Kda. Query: The cleaved nuclear fragment of SREBP1 is expected to be 60-70 Kda. But I'm getting the prominent band at 30Kda. Kindly help. While I would typically recommend loading between 20-40 ug of whole cell lysate or 10-15 ug of nuclear lysate per lane to ensure the maximum specificity, I do not think this could account for the bands you are detecting at incorrect molecular weights.
