The SPLASH protein also known as Synapse Protein LL Alpha SH3 functions as an adaptor in synaptic signaling. A molecular weight of approximately 50 kDa characterizes SPLASH and it shows high expression in neural tissues notably within the brain's synaptic regions. Synapse formation and plasticity strongly depend on the presence and function of this protein. It interacts with multiple synaptic components to assist in the organization of signal transduction complexes.
Biological function summary

The SPLASH protein coordinates with other synaptic proteins to facilitate neurotransmitter release and receptor clustering at the synapse. As part of larger protein complexes SPLASH forms dynamic interactions important for maintaining synaptic strength and plasticity. These complexes modulate synaptic vesicle exocytosis and endocytosis processes which are essential for efficient neuronal communication.

Pathways

The SPLASH protein integrates within the synaptic signaling pathway and the calcium signaling pathway. It serves as a link between the two pathways coordinating signal transduction events that modulate synaptic plasticity. SPLASH maintains functional connection with proteins like SNARE and PSD-95 which play central roles in synaptic vesicle fusion and postsynaptic density organization respectively.

The SPLASH protein links to neurodevelopmental disorders and synaptic dysfunction. It is particularly associated with conditions such as autism spectrum disorder where synaptic signaling and plasticity are affected. Moreover in Alzheimer's disease altered SPLASH expression influences amyloid precursor protein processing through its interactions with the Tau protein impacting cognitive function and neuronal integrity.

Secretory calcium-dependent phospholipase A2 that primarily targets extracellular lipids, exerting anti-inflammatory and immunosuppressive functions (PubMed : 10455175, PubMed : 10681567). Hydrolyzes the ester bond of the fatty acyl group attached at sn-2 position of phospholipids (phospholipase A2 activity) with preference for phosphatidylethanolamines and phosphatidylglycerols over phosphatidylcholines (PubMed : 10455175). In draining lymph nodes, selectively hydrolyzes diacyl and alkenyl forms of phosphatidylethanolamines, releasing omega-3 polyunsaturated fatty acids (PUFAs) such as eicosapentaenoate and docosahexaenoate that are precursors of the anti-inflammatory lipid mediators, resolvins (By similarity). During the resolution phase of acute inflammation drives docosahexaenoate-derived resolvin D1 synthesis, which suppresses dendritic cell activation and T-helper 1 immune response (By similarity). May act in an autocrine and paracrine manner (By similarity). Via a mechanism independent of its catalytic activity, promotes differentiation of regulatory T cells (Tregs) and participates in the maintenance of immune tolerance (By similarity). May contribute to lipid remodeling of cellular membranes and generation of lipid mediators involved in pathogen clearance. Displays bactericidal activity against Gram-positive bacteria by directly hydrolyzing phospholipids of the bacterial membrane (By similarity).
