Immunocytochemistry/ Immunofluorescence - Anti-Spermine antibody (AB26975)

ab26975 staining spermine in the Human HeLa cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with 4% formaldehyde for 15 minuts at room temperature (Upper images) or iced methanol of 3 minutes (Lower images) and blocked with 2% BSA for 30 minutes at 27°C. Samples were incubated with primary antibody (1/100) for 1 hour at 27°C. An Alexa Fluor®568-conjugated Donkey anti-rabbit polyclonal (1/1000) was used as the secondary antibody. Samples handled without ab26975 were used as negative controls (Left-hand panels).

Spermine is an organic polyamine compound with a molecular mass of 202.34 g/mol. It plays an essential role in cellular metabolism. Often found in eukaryotic cells spermine is also known as sym-norspermidine. The compound spreads across tissues with high concentrations in tissues that have rapid growth such as the thymus and testes. It exists intracellularly and consists of several amine groups which enable it to interact with negatively charged molecules like DNA and RNA. These interactions increase the stability and control of nucleic acids within the cellular environment.
Biological function summary

Spermine modulates various cellular processes. It is involved in cell growth and differentiation and it stabilizes cell membranes. By binding to DNA spermine helps preserve its structure during stress. Additionally spermine acts as an antioxidant reducing oxidative damage in the cells. Polyamines such as spermine often function independently rather than as part of a complex. The regulation of their synthesis and uptake is controlled by enzymes including ornithine decarboxylase which is an important player in polyamine biosynthesis.

Pathways

Spermine is central in the polyamine biosynthesis pathway which is vital for cellular proliferation. It regulates pathways involved in the synthesis and breakdown of proteins. The action of spermine interlinks with the metabolism of amino acids like ornithine and methionine influencing the biogenic amines pathway. Spermine's metabolic pathway intersects with proteins such as S-adenosylmethionine decarboxylase and spermidine synthase which closely control its levels in the cell.

Spermine has connections to several pathological conditions including cancer and neurodegenerative diseases. Its role in promoting cell growth links it to oncogenesis where abnormal regulation can lead to cancerous growths. Increased spermine levels associate with high-grade tumors. Likewise its oxidative stress modulation impacts neurodegenerative disorders like Parkinson's disease. The disruption in the polyamine pathway influencing spermine levels can exacerbate neuronal injury. Interactions with proteins such as N-methyl-D-aspartate (NMDA) receptors also highlight spermine's involvement in neurotoxicity.
