Spermidine and spermine are both polyamines found in living tissues and have various metabolic functions. As they are not proteins, they are not species specific. If the tissue you are interested in probing has spermine or speridine present the antibodies ab7318 and ab26975 should be able to detect the molecules.

The two antibodies which you are interested in (ab7318 and ab26975) are both formulations of the whole antiserum. This means that quantification of the exact concentration is not possible as there are several other proteins as well as the active antibody within the preparation. Typically, whole antiserum is of a concentration of 1-10 mg/ml.

I would suggest optimising the dilution of the antibodies for your experiment, starting with a dilution of 1/50-1/100. From there you should be able to see if you need to reduce or can increase the dilution needed for the optimum results. Ab26975 has not as yet been tested for its use in immunohisotchemistry, we would therefore be very interested in the results you obtain with this antibody. This antibody reacts with spermidine molecule and there is no differences between spermidine of different species. This antibody cannot be use to identify spermidine synthase.

The originator of this antibody has not provided details of how the western blot was performed. The epitope has not been mapped but as the molecule is very short we can't assume that there are multiples sites. This antibody was not characterized and tested in-house, but by the originator. We guarantee all applications and species reactivity stated on the datasheet. I do know that the polyamines (spermidine and spermine) are far too small to be detected by Western blotting. The antibody is used to detect polyamines incorporated in proteins rather than the polyamines themselves.
