    This antibody is used for the detection of LPS of Salmonella typhimurium.

    Salmonellae are Gram-negative , facultatively anaerobic bacteria of the family Enterobacteriaceae, made up of nonspore-forming rods, usually motile with flagella. Salmonella enterica serovar Typhimurium is the among the most common Salmonella serovars causing Salmonellosis food poisoning. S. typhimurium has a less alarming public image than S.typhi (which causes typhoid), but is a bigger health problem worldwide. Salmonellae possess 3 major antigens; the "H" or flagellar antigen (phase 1 & 2), the "O" or somatic antigen (part of the LPS moiety) and the "Vi" or capsular antigen (referred to as "K" in other Enterobacteriaceae). Salmonellae also possess the LPS endotoxin characteristic of Gram-negative bacteria. This LPS is composed of an "O" polysaccharide ("O" antigen) an "R" core and the endotoxic inner "Lipid A". Endotoxins evoke fever and can activate complement, kinin and clotting factors.

    Gels were (A) silver-stained or (B) Western blotted using the Anti-Salmonella typhimurium 0-4 antibody [1E6] (ab8274) as the primary antibody. Although equal amounts of cellular material were detected by silver staining, only the S. Typhimurium samples were recognized by the 1E6 antibody on the corresponding Western blot.

Unfortunately, we do not carry the biotinylated forms of these 2 antibodies, but we do carry biotinylation kits (e.g. https://www.abcam.com/EasyLink-Biotin-type-A-Conjugation-Kit-3-x-100µg-Biotin-ab102864.html - other sizes exist as well) if you like to biotinylate the antibodies on your own.

I have a question. I need an antibody that can bond to the body of Salmonella typhimurium VNP20009 and not its flagella. The bacteria are wanted to be able to swim after antibody conjugation, and that's why I don't want the antibody to get attached to the flagella. Do you have such a product?

We have 5 antibodies to Salmonella typhimurium:

ab69238 - was raised against the whole bacteria thus we do not know where the antibody would bind.

ab8274 and ab65922: both were raised against LPS of the bacterium which I suppose is located in the bacterial wall and not the flagellum - this should work (https://www.abcam.com/salmonella-typhimurium-0-4-antibody-1e6-ab8274.html)
(https://www.abcam.com/salmonella-typhimurium-lps-antibody-1e6-ab65922.html)

ab63901: was raised against the bacteria, but known to be reactive with the LPS - thus, it should work.

ab128709: raised against purified LPS from Salmonella typhimurium and reacts with an unidentified LPS determinant of Salmonella typhimurium - thus it should work for your purposes (https://www.abcam.com/Salmonella-typhimurium-LPS-antibody-1E6-ab128709.html).

Question

I am ready to submit an Abreview for the product listed below.
I tried to submit online, but there is no listing for Western Blot and no listing for Salmonella Typhimurium or Salmonella Enteritidis.
The 1E6 antibody worked very well on Western blots and was specific for Typhimurium.

After browsing your website, I had hard time to determine which pair of antibody (capture and detection) can work well on sandwich assays. Could you please recommend any antibody pair for sandwich assay for the detection of foodborne pathogens, especially Salmonella typhimurium, Listeria monocytogenes, or Campylobacter jejuni? Thank you for your enquiry regarding our Salmonella typhimurium, Listeria monocytogenes, or Campylobacter jejuni antibodies. For Listeria monocytogenes detection we recommend pair LZH1 (ab11439) – LZF7 (ab11440)(capture – conjugate) for sandwich immunoassay. (The best pair for sandwich ELISA was selected among the antibodies by testing all possible pairs). Unfortunately we have available only one antibody clone for the detection of Salmonella typhimurium. However, we are also offering antibodies for the detection of A, B and D-group specific salmonella O-antigens. We have information from a customer that they have used successfully the pair 10D9 (ab8272) – 5D12A (ab8273) (capture – conjugate) in immunochromatography for the detection of salmonella from food. The detection antibodies are available in conjugated form (e.g. hrp-conjugated). We are very sorry we do not have references for those antibodies. Here is the recommended protocol for Sandwich ELISA: Coating: Capture MAbs, 2.5 µg/ml in 0.1 M carbonate buffer, pH 9.3, overnight at + 4 C. Antigen: L. monocytogenes OM fraction or cell suspension, 1 h, 37 C. Conjugate: Conjugated MAbs in PBS-AT, 1 h, 37 C. Substrate: TMB, 15 min Using pair LZH1 – LZF7 the detection limits 0.3 ng/ml for L. monocytogenes OM fraction and 100 cells per well for L. monocytogenes cell suspension have been achieved. Question

I am doing research in detection of heat-killed Salmonella typhimurium by sandwich ELISA. I have some questions to choose the capture antibody from your products. How to decide what antibody to use? Will the antibody specificity be a lot different if I choose antibody produced from different animals, such as mouse, goat, rabbit ? Is it better to use a polyclonal antibody than monoclonal one if the sensitivity would be a limit for detection? I leant from your website about your products. The Salmonella typhimurium 0-4 antibody [1E6] (ab8274) and Salmonella typhimurium 0-4 antibody [8C11C] (ab8270) are the items I am interested in. What is the difference between these two antibodies? Can these antibodies bind with heat-killed Salmonella typhimurium and be used as solid phase capture antibody? Does it cross-react with other species? What is the specificity? Do you have biotin-conjugated anti-Salmonella antibody?

Both monoclonal antibodies recognize heat-killed Salmonella typhimurium. We have used the monoclonal antibodies (Mab) in ELISA according to the following scheme: antigen coating - Mab - Anti-mouse-IgG (conjugated), not in sandwich ELISA. The antibodies may be used for salmonella typhimurium detection in dot-blot, ELISA and immunofluorescence. Other applications have not been tested. Both clones do not recognise: S.paratyphi A, S.choleraesuis, S.newport, S. enteiditis, S. anatum, S. elandia, E.coli 055:B5, E.coli K12, Klebsiela pneum, Proteus vulgaris. Ab8274 has been shown to have a very broad cross-reactivity range recognising E.coli 1243 and Listeria monocytogenes species, being a potential positive control antibody for a variety of assays.
