RRAD also known as Ras-related associated with diabetes is a small GTPase belonging to the Ras superfamily. It weighs approximately 29 kDa and exhibits expression in cardiac and skeletal muscle tissues as well as the nervous system. This protein functions mechanically by binding and hydrolyzing GTP a process critical for its role in intracellular signaling. RRAD is known for its ability to affect various molecular interactions through its GTPase activity.
Biological function summary

RRAD regulates glucose transport and metabolism influencing energy balance in cells. It does not integrate into a larger protein complex but instead acts on its own to modify glucose transporter activity. RRAD impacts how cells take up and use glucose which is significant for maintaining cellular energy requirements and contributes to processes that control cell growth and survival.

Pathways

Understanding the signaling mechanisms of RRAD involves examining the insulin signaling pathway and the glucose metabolism pathway. RRAD impacts insulin receptor signaling and modulation of glucose transporter type 4 (GLUT4) activity. RRAD holds a relationship with other proteins such as AKT and PI3K in these pathways which play critical roles in transmitting signals that manage glucose uptake and regulation.

The alteration in RRAD expression or function associates with diabetes and cardiovascular diseases. In diabetes abnormal RRAD function affects insulin signaling contributing to insulin resistance. In cardiovascular disease RRAD dysregulation can influence cardiac function through its impact on energy metabolism. The protein interacts with others like GLUT4 and AKT which are relevant in these disease contexts implicating RRAD in pathways that influence disease progression.

May regulate basal voltage-dependent L-type Ca(2+) currents and be required for beta-adrenergic augmentation of Ca(2+) influx in cardiomyocytes, thereby regulating increases in heart rate and contractile force (By similarity). May play an important role in cardiac antiarrhythmia via the strong suppression of voltage-gated L-type Ca(2+) currents (By similarity). Regulates voltage-dependent L-type calcium channel subunit alpha-1C trafficking to the cell membrane (By similarity). Inhibits cardiac hypertrophy through the calmodulin-dependent kinase II (CaMKII) pathway (PubMed : 18056528). Inhibits phosphorylation and activation of CAMK2D (PubMed : 18056528).
