    actually, this time, I did incubation ON at 4°C with 1/1000 dilution Ab and it didn't work better. I'm gonna try with 2 fold increase of Ab concentration but I'm pretty sure it is not gonna be better because I don't have the faintest band with a 1/1000 dilution. About the ab56560 I'm aware of unspecific bands but anyway I want to give a shot with this antibody because unspecific bands may depend on cell lines. Could you send me it ?

    Please let me know if increasing the concentration does not prove to be successful. If it does not help, I would be happy to send you ab56560 as a replacement, so that you can try and get a signal in our western blot.     I received another lot of the ab83179 antibody against RNAseH1 you shipped me after I experienced trouble with the first lot. Unfortunatly I didn't get any signal in WB although, as a positive control, I can detect very easily on the same membrane RNAseH2A with the ab83943 antibody (from rabbit also). I tried to detect RNAseH1 on 3 cell lines : Huh7, Hela and 293T. I used the recommended concentration of ab (1ug/mL) and block with milk. Last time, you proposed me to switch to the ab56560 antibody against RNAseH1. Can you thus send me a tube of this ab ?

    Thank you for your reply. I am sorry that the replacement antibody that you received failed to work in western blot. I was wondering if with the replacement antibody you tried an overnight incubation of the primary antibody. I only ask, as I remember that during our previous troubleshooting emails, it was something we had discussed but not tried. Also, I know you were trying the antibody concentration of 1ug/ml, it may be worth increasing the concentration to 2ug/ml (1/500 dilution). It could be that you need to use the antibody at higher concentration and incubate for longer. I also remember that during our previous emails, you raised concerns about the presence of multiple bands in the western blot image shown for ab56560. Therefore I am concerned that if you were to use this antibody and you receive multiple bands, the antibody may still not be suitable.  I just wanted to mention this and see if you would still want to try ab56560. The alternative is that I could refund the original purchase for ab56560. I look forward to your reply.

    I already use the recommended concentration of this ab (ab83179) : 1ug/mL and I do block in 5% milk. I agree that overnight incubation at 4°C may enhance detection for weak signal but the signal is not weak, there is no signal at all ! Could you send me another tube of ab83179 from another lot ?

    Hi, I bought 2 antibodies from your compagny two weeks ago : anti RNAseH1 (ab83179) anti RNAseH2A (ab83943) the anti RNAseH2A works very good with a band at the expected size (see pp). On the contrary, the anti RNAseH1 gave no signal, even not a faint band with overexposure (see pp). I did the WB twice with the same result with two different cell lines. I sent the scanned film of the WB as attached file. What do you propose for your customer in that case ?

    Thank you for contacting Abcam. I am sorry that you are experiencing problems with ab83179 in western blotting. I have some questions/suggestions about the protocol that you are using. 1 - What concentration of ab83179 are you using? It has been tested at 1ug/ml, so depending on the concentration you are using, increasing the concentration could help. 2 - Incubating overnight at 4C is also recommended for antibodies that gave weak signal. 3 - Are you using 5% non-fat dried milk as your blocking agent? The antibody ab83179 is covered under our Abpromise and so is guaranteed to work in western blot on human samples. If we cannot resolve this issue then I would be happy to send you are replacement antibody or process a refund. I look forward to your reply.
