  • 功能

    Regulates endocytic recycling. May exert its functions by interacting with multiple effector proteins in different complexes. Acts as a major regulator of membrane delivery during cytokinesis. Together with MYO5B and RAB8A participates in epithelial cell polarization. Together with RAB3IP, RAB8A, the exocyst complex, PARD3, PRKCI, ANXA2, CDC42 and DNMBP promotes transcytosis of PODXL to the apical membrane initiation sites (AMIS), apical surface formation and lumenogenesis (By similarity). Together with MYO5B participates in CFTR trafficking to the plasma membrane and TF (Transferrin) recycling in nonpolarized cells. Required in a complex with MYO5B and RAB11FIP2 for the transport of NPC1L1 to the plasma membrane. Participates in the sorting and basolateral transport of CDH1 from the Golgi apparatus to the plasma membrane. Regulates the recycling of FCGRT (receptor of Fc region of monomeric Ig G) to basolateral membranes.

    BATCH NUMBER 56526 ORDER NUMBER 130916 DESCRIPTION OF THE PROBLEM No band SAMPLE Cell extract PRIMARY ANTIBODY Rab11A (ab 11034) 1:100 up to 1:1000 in blocking buffer, incubated overnight, washed 3x 5min in PBS DETECTION METHOD ECL, exposure time 0,5s to 30min POSITIVE AND NEGATIVE CONTROLS USED No primary ANTIBODY STORAGE CONDITIONS -20?C SAMPLE PREPARATION Protein extracts were prepared in Tris buffer with 1% Triton X-100, NaCl, EGTA, EDTA, and 0,5% NP40 supplemented with protease inhibitor cocktail. The lysates were aliquoted and kept in -80?C. AMOUNT OF PROTEIN LOADED 50ug-150ug ELECTROPHORESIS/GEL CONDITIONS 10-12% reducing gel TRANSFER AND BLOCKING CONDITIONS 60 min up to 90 min wet transfer containing 20% methanol. Blocking with 5% milk in PBS 60 min. SECONDARY ANTIBODY anti mouse HRP conjugated 1:3000 to 1:10000 HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 6 HAVE YOU RUN A "NO PRIMARY" CONTROL? Yes DO YOU OBTAIN THE SAME RESULTS EVERY TIME? Yes WHAT STEPS HAVE YOU ALTERED? None ADDITIONAL NOTES The protcol described here has been used for at least 10 other antibodies among them several are from abcam but we have problems only with the Rab11A ab11034 antibody. I am absolutely sure that there is nothing wrong with the protocol. I would really appreciate if you could me send an answer as soon as possible otherwise I will have to order this from another company and finish this last experiment for our manuscript. It is quite urgent.
