Western blot : Anti-PYCR2 antibody ab103535 staining at 1/500 dilution, shown in green; Mouse anti alpha Tubulin ab7291 loading control staining at 1/20000 dilution, shown in magenta. A band was observed at 34 kDa in Wild-type MCF7 cell lysates with no signal observed at this size in PYCR2 knockout MCF7 cell line. To generate this image, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 5pc Milk in TBS-0.1 % Tween 20 (TBS-T) before incubation with primary antibodies overnight at 4 C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit 800CW and Goat anti-Mouse 680RD at 1/20,000 dilution.

PYCR2 also known as Pyrroline-5-carboxylate reductase 2 is an enzyme involved in proline biosynthesis. It has a molecular mass of approximately 31 kDa. This protein catalyzes the conversion of pyrroline-5-carboxylate to proline an amino acid important for protein synthesis and cellular stress responses. Expressed broadly in human tissues PYCR2 shows notable expression in the liver brain and various muscle tissues indicating its wide functional relevance across different physiological systems.
Biological function summary

The enzyme participates in maintaining proline levels and contributes to cellular osmotic balance and protection against oxidative stress. It is not typically part of a larger protein complex but interacts with other metabolic enzymes that support similar stress response pathways. By catalyzing an important step in the proline biosynthesis pathway PYCR2 helps sustain cellular redox states and protein turnover rates under stressful conditions.

Pathways

PYCR2 plays a significant role in the proline metabolism pathway and glutamate pathway. The functioning of this enzyme in these pathways supports the synthesis and recycling of proline which is important for cell survival under osmotic and oxidative stress conditions. The disruptions in PYCR2 activity may also alter interactions with proteins involved in these pathways such as Δ1-Pyrroline-5-carboxylate synthase thereby exacerbating the disease states.

At physiologic concentrations, has higher specific activity in the presence of NADH (PubMed : 23024808, PubMed : 2722838, PubMed : 6894153). Involved in cellular response to oxidative stress (PubMed : 25865492).
