Immunocytochemistry/ Immunofluorescence - Anti-PUF60/FIR antibody (AB22819)
ICC/IF image of ab22819 stained MCF7 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal donkey serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab22819, 5μg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 donkey anti-goat IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43μM.

Biological function summary
PUF60/FIR is essential for mRNA processing. It is part of the spliceosome complex where it interacts with other splicing factors to facilitate pre-mRNA splicing. PUF60/FIR regulates alternative splicing events which can influence gene expression profiles enabling response to different physiological conditions. It connects with the regulation of cell growth and differentiation through its splicing activities.
Pathways
PUF60/FIR participates in the regulation of transcription and mRNA processing pathways. These pathways include the transcription regulation of MYC a critical protein for cell proliferation and apoptosis. PUF60/FIR modulates the splicing and transcription of RNA polymerase II-dependent genes functioning in interaction with other proteins such as SF3B1 and hnRNP H. This interaction influences the transcriptional activity of gene networks involved in cell cycle progression and survival pathways.
