Protein Tyrosine Phosphatase Alpha (PTP alpha) also known as PTPRA serves as an enzyme that removes phosphate groups from specific tyrosine residues on proteins. It has an approximate molecular weight of 130 kDa. PTP alpha is located in the cell membrane and shows expression in various tissues including the brain liver and skeletal muscle. It functions as a signaling molecule that modifies the activity of several proteins playing a significant role in cell growth differentiation and mitosis.
Biological function summary

PTPRA influences cellular processes by serving as a regulator of the Src family of tyrosine kinases which are important for intracellular signal transduction. PTP alpha does not form part of a complex but rather exists as a monomer although it can interact with other proteins. Its activity helps maintain the balance of phosphorylation and dephosphorylation which is essential for cellular signaling homeostasis.

Pathways

PTP alpha acts in the integrin and growth factor receptor signaling pathways. It plays a role in integrin signaling by dephosphorylating substrates that modulate cell adhesion and migration. Additionally in growth factor receptor signaling PTP alpha modulates cell proliferation by interacting with substrates such as the epidermal growth factor receptor (EGFR). It shares pathway links with proteins like SRC and FAK emphasizing its regulatory role over them.

PTP alpha has associations with cancer particularly in cases such as colorectal and breast cancer. It affects cancer progression by modifying signaling involved in uncontrolled cell growth. Furthermore PTPRA has findings connecting it with neurodegenerative diseases like Alzheimer's through its effect on signaling cascades that involve amyloid precursor protein (APP). In these contexts PTP alpha’s interactions with proteins such as EGFR and FAK reveal its significance in disease pathology.

Following integrin engagement, specifically recruits BCAR3, BCAR1 and CRK to focal adhesions thereby promoting SRC-mediated phosphorylation of BRAC1 and the subsequent activation of PAK and small GTPase RAC1 and CDC42 (By similarity).
