The antibody was purified from rabbit serum by sequential epitope-specific chromatography. The antibody has been negatively preadsorbed using a non-phosphopeptide corresponding to the site of phosphorylation to remove antibody that is reactive with non-phosphorylated Inhibitor-2. The final product is generated by affinity chromatography using an Inhibitor-2 -derived peptide that is phosphorylated at threonine 72. -

Immunocytochemistry/ Immunofluorescence - Anti-Protein phosphatase 1 inhibitor subunit 2 (phospho T72) antibody (ab27850)Immunocytochemistry/Immunofluorescent analysis of A431 cells treated with Nocodazole for 24 hours, staining Protein phosphatase 1 inhibitor subunit 2 at 1/250 dilution, followed by Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate at 1/2000 dilution (green). Cells were fixed with 4% paraformaldehyde and permeabilized with 0.1% Triton X 100 for ten minutes, and blocked with 1% BSA for one hour at room temperature. Nuclei were counterstained with DAPI (blue). F-actin was stained with Rhodamine Phalloidin (red).
Negative controls are untreated A431 cells (e) and a secondary only antibody control (f).

Cells were grown on a chamber slide and fixed in cold 10% trichloroacetic acid for 10 minutes, rinsed in TBS and permeabilized with 0.1% Triton-X-100 for 15 minutes.
