    Prolactin (PRL) is a hormone involved in a variety of important functions including ion transport and osmoregulation, stimulation of milk, protein synthesis as well as the regulation of numerous reproductive functions. PRL exerts its influence on different cell types through a signal transduction pathway which begins with the binding of the hormone to a transmembrane PRL receptor. Immunoreactive PRL receptor, a member of the cytokine receptor family, varies in size (short and long forms) with tissue source and species, from ~40 kDa to 100 kDa. The PRL receptor consists of at least three separate domains: an extracellular region with 5 cysteines which contains the prolactin binding site, a single transmembrane domain and a cytoplasmic region, the length of which appears to influence ligand binding and regulate cellular function.

      Mammary gland tissue from lactating light horse mare was incubated with ab2772 at 1:100 for 1 hour at 23°C and for 24 hours at 4°C. A biotin-conjugated goat anti-mouse IgG was used as the secondary antibody (23°C for 1 hour) .Development using avidin-biotin complex 23°C for 1 hour plus 3,3-diaminobenzadine for 15 min and counterstained with haematoxylin. Antigen retrieval was facilitated using steaming at 95°C for 20 min in Trizma® buffer (pH 10.0) . Blocking endogenous peroxidase performed with 0.3% H2O2 and MeOH (20 min at 23°C). To decrease non-specific staining, tissue was pre-incubated in normal goat serum for 30 min at 23°C.

Cells were probed without (control) or with ab2772 (1:20) over night at 4°C, washed with PBS and incubated with a DyLight-488 conjugated secondary antibody. Images were taken at 60X magnification.

      Overlay histogram showing MCF-7 cells stained with ab2772 (red line). The cells were fixed with 80% methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab2772, 2µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

      ab2772 staining the Prolactin Receptor/PRL-R in Human salivary gland tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with paraformaldehyde and blocked with 10% serum for 1 hour at 20°C; antigen retrieval was not performed. Samples were incubated with primary antibody (1/100) for 12 hours at 4°C. A Biotin-conjugated Rabbit anti-mouse polyclonal (1/200) was used as the secondary antibody.

    Hello,
    Thank you for your answer. I understand your concern about the possible non-specific signal. However, we usually use other mouse antibodies on prostate IHC of mouse and we hardly see any non-specific signal and always localized to the extracellular matrix surrounding the glands and/or the lumen. We don't see non-specific signal in the epithelium. So, my question is: will it be ok if I try the antibody in our usual conditions or do you require I use the mouse blocking system to get the discount?

    Question

    Hello,
    I
    would like to test the Anti-Prolactin Receptor antibody [U5] (ab2772)
    in IHC of paraffin-embedded mouse prostate tissue. According to the
    datasheet, this antibody recognizes rat, rabbit, human, and pig PRLR,
    but it hasn't been tested on mouse. Could you let me know if I would get
    the 100% testing discount if I do the test and submit the abreview?
    Also, I welcome any suggestions for the tests that might not be included
    in the datasheets. Or suggestions of other antibodies that might be more
    suitable for my needs.

    Thank you for your enquiry and your interest in our product.

    We have already offered a testing discount for one of our customers who has tested ab2772 on mouse skin tissue sections. The results have been published as an Abreview and the image can be seen on the on-line datasheet. Since the rating of this feedback was only 3 star (and not 5 star), we are not convinced that ab2772 is suitable for detecting murine Prolactin Receptor antibody and have not updated the tested species section with mouse. We would still welcome more results from other customers.

    Looking at this particular image, it seems that the background signal is pretty high. Since this primary antibody is raised in mouse and the samples are from the same species, it is likely that the antibody may recognize some endogenous non-specific immunoglobulins.

    I could certainly offer you a discount code for testing but it would be worth perhaps considering using a mouse-blocking to reduce any potential high background signal.

    Thank you for your email. As a positive control I can recommend to use sections from breast, placenta, kidney, liveror pancreas. Indeed, the prolactin receptor is expressed in those tissues. (http://www.uniprot.org/uniprot/P16471) There should however also be an expression in the salivary gland, as seen on the datasheet of ab2772. We do have in our catalogue tissue slides which can be used as positive controls. As shown on the Abreviews (feedback obtained from end users), the prolactin receptor antibody ab2772 has been successfully used in IHC-Fr on Mouse skin sections and in IHC-P on Human salivary gland sections fixed with paraformaldehyde : https://www.abcam.com/index.html?pageconfig=reviews&intAbID=2772 . For IHC-P, the antibody was used at a dilution 1/100, for 12 hours at 4°C. No antigen retrieval method was applied. Another positive control recommended for alternative prolactin receptor antibodies is breast carcinoma. I hope this information is helpful to you.     Do you have an antibody that will recognize the long form of the protein, and a positive control tissue for the long form? I have tried ab2772 on various tissues from mouse (liver) and human (breast carcinoma, lung) but only see the short form at 40kD.

    The following references cite detection of the long and short Prolactin receptor proteins using clone U5 (though not specifically Abcam's ab2772): Short Form (~40 kDa) AJP Endocrinol Metab Vol 287, E750-E757 Sept 2004. http://ajpendo.physiology.org/cgi/reprint/287/4/E750 Detects short PRLR from mouse total liver lysates. Long Form (~100 kDa) J. Biol. Chem., 270(10):5136-5143, 1995. http://www.jbc.org/cgi/content/full/270/10/5136 Detects long PRLR from rabbit mammary gland membranes.     Ab2773 has been shown to be specific for the short form around 42 kDa. The other Prolactin Receptor antibody that we currently have, ab2772, detects both the short and long forms. So unfortunately we don't have what your customer is looking for.
