In Western blot, ab109441 was shown to bind specifically to Plakophilin 3. A band was observed at 70 and 80 kDa in wild-type HeLa cell lysates with no signal observed at this size in PKP3 CRISPR-Cas9 edited cell line ab265539 (CRISPR-Cas9 edited cell lysate ab258120). The band observed in the CRISPR-Cas9 edited lysate lane below 70 and 80 kDa is likely to represent a truncated form of Plakophilin 3. This has not been investigated further and the functional properties of the gene product have not been determined. To generate this image, wild-type and PKP3 CRISPR-Cas9 edited HeLa cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged.Secondary antibodies used were Goat anti-Rabbit IgG H&L (IRDye®  800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) at 1/20000 dilution.

    In what period of time is recommendable to use the antibody once it is defrosted and opened?

    Thank you for contacting us.

    We would not recommend to freeze the antibody again once it has been thawed and used on a few samples. These repeated freeze/thaw cycles could damage the antibody. Instead, we would suggest to prepare and freeze small aliquots of the antibody. In case you have already frozen the whole vial before aliquotting, it should be alright to defrost it, prepare the aliquots and freeze them once again. Just keep in mind that any reduced signal might be caused by this.

    Once the antibody (an aliquot) has been defrosted it can be kept at 4ºC for a few weeks. Here, we would recommend to store the stock solution and preparedilutions freshly when required.

    I can confirm that ab109441 has been tested successfully in IHC-P at a dilution of 1/100 - 1/250, as stated on the datasheet. Please note this is a guideline only, and may require some further optimization.

    For a small section of tissue, you can pipette 50 ul of diluted antibody solution ontoone slide.For larger sections, you may require 100 - 200 ul per slide. As long as thesolution covers the section and does not dry out during incubation.

    Unfortunately, I am sorry this antibody is currently out of stock. The expected wait time is 2 - 3 weeks to delivery. This information can also be found on the datasheet. I can suggest to select the correct country in the top right hand corner of the screen to see the correct information:
