    This is a calcium-independent, phospholipid-dependent, serine- and threonine-specific enzyme. Phosphorylates 'Ser-311' of RELA subunit of NF-kappa-B.
    PKC is activated by diacylglycerol which in turn phosphorylates a range of cellular proteins. PKC also serves as the receptor for phorbol esters, a class of tumor promoters. Subunit of a quaternary complex that plays a central role in epithelial cell polarization.

Question

I am a bit concerned about the discrepancy in molecular weights between this phospho-PKCzeta (Thr560) antibody, which detects a band at around 30kD, and the whole PKC-zeta antibodies offered by Abcam which detect bands at around 75kD (predicted molecular weight for PKC-zeta is about 70kD).

Can you explain why this phospho antibody is detecting a band at 30kD? I use a phospho-PKC antibody right now (Thr 410) that detects a band where it is expected, up around the 70kD region, near where the whole PKCzeta protein runs.

What is your explanation for this?

Question

Dear Madam/Sir,   I have recently read an interesting article regarding the regulatory role of PKCzeta isoform on F0F1 ATP synthase beta subunit. It claimed that PKCzeta is able to phosphorilate the beta subunit of the F0F1 ATP synthase and hence, regulate its activity. The phosphorilation was shown by Western blot using antibodies raised against the phospho- and the nonphospho forms of the synthase. Unfortunately, it was not written who was the provider of the antibodies, and the authors didn't reply my e-mails to tell it to me.    Finally, I have checked the homepage of MitoSciences, but I failed to find antibodies against the phospho form of the ATP syntase, so my question is: could you provide me antibodies raised against both the phospho and the nonphospho forms of the beta subunit of human F0F1 ATP synthase? Or alternatively, could you suggest any methods with which I could measure somehow the activity of the ATP synthase?   Thank you in advance!
