      IHC image of ab23705 staining in human pancreas formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab23705, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    Das Labor, das Anti-Phospholipase A2Antikoerper ab23705 herstellt, hat bestätigt, dass das Immunogen das Volllängenprotein sPLA2 (human Type IIA) mit der SwissProtID P14555.2 ist. Damit ist dieser Antikörper spezifisch für die Membran-assoziierte PLA2.

    Die Antikörper ab124230 und ab73406 sind hingegen spezifisch für die cytosolische PLA2 mit der SwissProtID P47712.

    Da diese beiden Formen sehr unterschiedlich sind, haben wir leider keinen Antikörper verfügbar, der beide Formen erkennt.

    Do you have an in-house support for the use of this antibody in various formalin-fixed paraffin-embedded tissues? I am particularly interested in the brain. Thanks. I have searched the literature and found several articles for your antibody success in pancreas and aorta and mycoardium, as well as in gastric cancer. A competitor's product was tested in-house and had high specificity for many tissues but they were able to tell me beforehand that it did not stain in the brain. I know that your antibody is guaranteed to work or our money back, but I would like some additional support for success in our use before ordering the antibody, if that information was available.

    Hi. I have another question and no one local to tap their expertise in IHC. In your technical bulletin for IHC protocols you are using Tris/Triton X-100 for permeabilization/washes. I have been using a Tween20/Tris buffer for my washes. One article using another vendor's sPLA2 used PBS/Triton X-100 (and allowed the sample to incubate for 30 minutes in this solution following H202 endogenous peroxide blocking) for permeabilization. Most articles do not even include what buffer they are using for their washes in their materials and methods. In your experience does the choice of buffer make a difference in the success of the detection of the antigen of interest? I did find it interesting that you suggested the H202 blocking after the incubation of the primary antibody if using the HRP system and I will be giving that a try. I really do appreciate your quick responses.

    Many of the protocols out there will call for "either Triton-X or Tween20" for permeabilization/washes and I think either would be a good choice. The advantage to Tween20 is that it is more gentle and less likely to disrupt the cell, however Triton-X is more fast acting. I hope this information helps.     Hi. I am inquiring about the sPLA2 rabbit poly-clonal antibody and its use in IHC-P. I have attempted numerous studies on human brain tissues using sPLA2 antibodies from other vendors and have not had consistent results and am considering trying AbCam. There is one published article from China in which they uses the antibody on rats pancreas but did not give details on which antigen retrieval buffer they used nor the source of their secondary antibody. Additionally, they incubated at 37C for all three steps. Your data information sheet is from formalin-fixed human tissue, using citrate buffer for AR, a short primary antibody incubation step at room temperature, but the secondary/tertiary reagents are not typical of what I have available. I will be using a manual LSAB system (we are a laboratory with somewhat limited funds at the moment), Tris/Tween buffer wash (I noticed that your standard protocol calls for Triton X) with DAB as the chromagen. I have read your information on IHC protocols but not sure what the data sheet for ab23705 is referring to for "protocol F". Do you think I will have success with your antibody or will I need to move up to a more sensitive detection system? There is not a lot of published results for sPLA2 IHC on human formalin-fixed tissues for me to ascertain whether my normal IHC protocol will be successful. I usually go overnight with the primary antibody at either 4C or room temperature. I would appreciate additional information on the HRP conjugated compact polymer system that you utilized in your studies for development of the antibody. Thanks much for any suggestions that you may have as I try to convince my PIs that we need to consider a new source for the sPLA2 antibody.
