ELISA - Anti-PGC1 alpha + beta antibody (AB72230)

ab72230 was tested using an Indirect ELISA approach. The wells were coated with peptide (1μg/ml at 100μl/well) overnight at 4°C, followed by a 5% BSA blocking step for 1 hour at room temperature. The primary Ab was then added at a dilution range of 1- 0.00025μg/ml (100μl/well) for 1hr at room temperature. A HRP-conjugated anti-rabbit IgG (heavy and light chain) was used as a secondary antibody at 1 : 20,000 dilution for 1hr at room temperature.

ab72230 gave a positive result in ELISA against PGC1 alpha peptide and PGC1 beta peptide. This indicates that it recognises both forms.

The immunogen design for ab72230 is shown to cross-react with both PGC1 alpha (ID: 124782 and Swissprot Q9UBK2) and PGC1 beta (ID: 240776 and SwissProt Q86YN6). From Jan 2024, QC testing of replenishment batches of this polyclonal changed. All tested and expected application and reactive species combinations are still covered by our Abcam product promise. PGC1 alpha and beta also known as Peroxisome Proliferator-Activated Receptor Gamma Coactivator 1-alpha (PGC-1α) and 1-beta (PGC-1β) are transcriptional coactivators with molecular weights of approximately 91 kDa and 102 kDa respectively. These proteins are important in regulating energy metabolism and are highly expressed in tissues with high energy demand such as skeletal muscle heart and brown adipose tissue. The proteins interact with nuclear hormone receptors and various transcription factors to control the expression of genes involved in energy homeostasis.
Biological function summary

PGC1 alpha and beta activate and coordinate the expression of genes involved in mitochondrial biogenesis and oxidative phosphorylation.
