Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-PDE6 alpha antibody (AB5659)
IHC image of ab5659 staining in human renal carcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab5659, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Biological function summary
PDE6 alpha functions within a larger enzyme complex known as the PDE6 holoenzyme. This complex consists of multiple subunits including the PDE6 beta and PDE6 gamma subunits which form the catalytic core. PDE6 alpha contributes to the regulation of cGMP levels which is essential for maintaining the electrical response of photoreceptors to light. The precise action of hydrolyzing cGMP mediates the phototransduction cascade that eventually leads to visual perception.
Pathways
With regard to pathways in cellular function PDE6 alpha is an important player in the phototransduction and visual signal transduction pathways. Within the phototransduction pathway PDE6 alpha interacts closely with other proteins like rhodopsin and transducin. These proteins activate PDE6 alpha upon light exposure initiating the cascade that results in hyperpolarization of the photoreceptor cell membrane. The role of PDE6 alpha in managing cGMP concentration positions it as a major component in these pathways.
