    Defects in PAX8 are the cause of congenital hypothyroidism non-goitrous type 2 (CHNG2) [MIM:218700]. CHNG2 is a disease characterized by thyroid dysgenesis, the most frequent cause of congenital hypothyroidism, accounting for 85% of case. The thyroid gland can be completely absent (athyreosis), ectopically located and/or severely hypoplastic. Ectopic thyroid gland is the most frequent malformation, with thyroid tissue being found most often at the base of the tongue.

    1) How was antibody specificity tested?

    -We have done ELISA, Western Blot, and IHC for the specificity tests.


    2) Do you have any information regarding cross-reactivity of this Pax8 antibody with other Pax-Proteins, especially Pax2 and Pax5?

    -BC12 does not cross-react with other PAX family members, like Pax2 and Pax5.



    3) Do you provide detailed information on the immunogen

    -recombinant human PAX8



    4) What is the epitope recognized by the antibody?

    -Epitope: aa258-272 of human PAX8.

    The ab124445was raised by immunizing full length of human PAX8. The epitope maps to an internal sequence. There is no cross-reactivity with PAX2 and PAX5.

    The immunogen isthe recombinantfull-length human PAX8 protein. The exact epitope has not been mapped, therefore we do not know where the antibody would bind in this region.

    As for the specificity and cross-reactivity with Pax2 and Pax5, I will check with the lab for potential data or additional information.
