      Polymerase that creates the 3'-poly(A) tail of mRNA's. Also required for the endoribonucleolytic cleavage reaction at some polyadenylation sites. May acquire specificity through interaction with a cleavage and polyadenylation specificity factor (CPSF) at its C-terminus.

      Polysumoylated. Varying sumolyation depending on tissue- and cell-type. Highly sumoylated in bladder and NIH 3T3 cells. Sumoylation is required for nuclear localization and enhances PAP stability. Desumoylated by SENP1. Inhibits polymerase activity.
      Hyperphosphorylation on multiple CDK2 consensus and non-consensus sites in the C-terminal Ser/Thr-rich region represses PAP activity in late M-phase. Phosphorylation/dephosphorylation may regulate the interaction between PAP and CPSF.
      Acetylated in the C-terminus. Acetylation decreases interaction with NUDT21 and KPNB1, and inhibits nuclear localization through inhibiting binding to the importin alpha/beta complex.

      Question

      1) Antibody dilution was 1:2000 in one experiment and 1:1000 in the second. The dilution buffer contained 5% BSA and 0.4% sodium azide in TBST.

      2) The membrane was blocked for 30 minutes in 5% milk in TBST.

      3) The blot was completely empty. I did Ponceau stain the membrane and there were no problems with the transfer.

      4) Yes, the secondary is HRP-conjugated goat anti-rabbit antibody. We know it works because we use it all the time for other antibodies.

      5) The positive control antibody was to beta-actin. It is a mouse monoclonal we purchased from Abcam (ab6276).

      I don't mind a replacement if you think that this is a batch-specific problem and that this antibody generally works. Let me know if you have any additional questions.

      Thank you for the protocol and result details. Regarding a replacement, there have not been other complaints about the antibody but not many people have purchased it. We have only the lot that you received, and I am concerned that another vial will simply give you the same result. I think a better choice for a replacement is ab126934:

      Protocol: I performed two independent western blots using varying amounts of cell lysate from HEK293 cells. I probed with both anti-PAPOLA (first), and a positive control antibody (second). In both experiments, only the positive control antibody gave any signal.

      2) What was used for blocking?
      3) You reported no signal: Did you see any other bands, or was the blot completely empty?
      4) I assume the secondary antibody was HRP-conjugated, and is working well otherwise?
      5) What was your positive control antibody?

      The options weoffer are: a free of charge replacement with another PAPOLA antibody or an antibody of your choice (e.g. to a different target protein), a credit or a refund.
