Immunocytochemistry/ Immunofluorescence - Anti-pan Synuclein antibody (AB6176)

ICC/IF image of ab6176 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab6176, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Pan Synuclein also known as alpha beta and gamma synucleins are small proteins with molecular masses around 14-19 kDa. These proteins are expressed largely in the central nervous system and are also present in other tissues such as the peripheral nervous system and certain types of muscles. They share structural similarities and comprise a family often involved with neurons where they play roles in various cellular processes.
Biological function summary

Synucleins participate in the regulation of synaptic function and plasticity influencing neurotransmitter release and membrane dynamics. They do not form a specific complex but can interact with lipid membranes and various cellular proteins affecting vesicular transport processes. These proteins hold influence over essential cellular functions that depend on synaptic activity and efficiency.

Neuronal protein that plays several roles in synaptic activity such as regulation of synaptic vesicle trafficking and subsequent neurotransmitter release (PubMed : 20798282, PubMed : 26442590, PubMed : 28288128, PubMed : 30404828). Participates as a monomer in synaptic vesicle exocytosis by enhancing vesicle priming, fusion and dilation of exocytotic fusion pores (PubMed : 28288128, PubMed : 30404828). Mechanistically, acts by increasing local Ca(2+) release from microdomains which is essential for the enhancement of ATP-induced exocytosis (PubMed : 30404828). Also acts as a molecular chaperone in its multimeric membrane-bound state, assisting in the folding of synaptic fusion components called SNAREs (Soluble NSF Attachment Protein REceptors) at presynaptic plasma membrane in conjunction with cysteine string protein-alpha/DNAJC5 (PubMed : 20798282). This chaperone activity is important to sustain normal SNARE-complex assembly during aging (PubMed : 20798282). Also plays a role in the regulation of the dopamine neurotransmission by associating with the dopamine transporter (DAT1) and thereby modulating its activity (PubMed : 26442590).
